摘  要:目的:比较采用电穿孔和脂质体lipofectamine两种方法转染人肝癌细胞的效率优劣。方法:分别采用电穿孔和脂质体转染miR-218进入人肝癌细胞Hep G2,采用RT-PCR、Western blot、流式细胞仪、Tunel检测等方法比较两种方法的转染后对人肝癌细胞的影响。结果:采用两种方法都可以成功将miR-218转染进人肝癌细胞Hep G2中,降低了Hep G2中miR-218靶基因RNA编辑酶ADAR1的表达,使得细胞凋亡增加,但电穿孔转染组效率更高。结论:电穿孔转染是一种安全、高效的转染方法,值得推广。Objective: To compare the transfection efficiencies of miR -218 into human hepatocarcinoma cells HepG2 by electroporation or lipofectamine. Methods: miR- 218 was transfected either by electroporation or by lipo- fectamine. Cells were collected, and then the condition of HepG2 cells were detected by RT - PCR, Western blot, Flow Cytometry and Tune1 test. Results:Both two methods could transfect miR -218 into HepG2 swimmingly,leading to the increase of apoptosis ,with the apporach of electroporation had abetter effect. Conclusion:Electroporation transfection is a safe. effective approach, which is worthy oromotion.
