摘 要:目的:阿苯达唑(Albendazole,ABZ)是一种苯并咪唑类广谱抗寄生虫药物,已在多种癌细胞中证实具有抗肿瘤作用,但其对胃癌细胞的作用鲜有报道。本文探讨其对人胃癌细胞MKN-45的迁移、侵袭能力的影响以及可能的分子机制。方法:用不同浓度ABZ处理人胃癌MKN-45细胞,采用细胞划痕实验,Transwell小室实验检测细胞的迁移侵袭能力,Western blot法检测MKN-45细胞中E-cadherin和MMP-9蛋白的表达水平,ELISA法检测细胞VEGF分泌水平。结果:阿苯达唑作用于人胃癌细胞MKN-45后,细胞划痕愈合的程度及Transwell穿膜细胞数随浓度增加有不同程度的改变,0.25、0.5μmol/L ABZ可显著抑制细胞迁移率(F=39.311,P<0.01),减少迁移细胞数(F=11.331,P<0.01)及侵袭细胞数(F=7.671,P<0.01);经ABZ处理的MKN-45细胞E-cadherin蛋白表达水平上调而MMP-9蛋白表达水平明显下调,VEGF分泌水平明显下降。结论:阿苯达唑可能通过上调E-cadherin及下调MMP-9表达水平来抑制胃癌细胞的迁移及侵袭能力,并能抑制胃癌细胞VEGF的分泌。Objective:To determine the effects of Albendazole(ABZ) on migration and invasion of human gastric cancer cell lines MKN - 45, as well as the possible mechanisms. Methods : Scratching assay and transwell method were used to measure the effects of ABZ on the migration and invasion/metastasis abilities of MKN - 45 cells in vitro. The changes of E - cadherin and MMP - 9 expression after Albendazole treatment in MKN - 45 cells were surveyed by Western blot. Production of vascular endothelial growth factor (VEGF) was detected by ELISA. Results: Treatment with ABZ at a dose of 0.25 and 0.5 p, mol/L significantly inhibited the migration rate ( F = 39.311, P 〈 0.01 ), de- creased the migration number ( F = 11.331, P 〈 0.01 ) and invasion number ( F = 7.671, P 〈 0. 1 ). ABZ treatments significantly reduced the protein levels of MMP - 9 and VEGF, but increased the protein levels of E - cadherin. Con- clusion: ABZ can inhibit the migration and invasion of gastric cancer cells by up - regulating E - cadherin and down - regulating MMP - 9 expression levels, and can suppress the secretion of VEGF in gastric cancer cells.
