摘 要:探索了覆盆子总酚酸的大孔树脂纯化工艺。以总酚酸的吸附率与解吸率为考察指标,通过单因素和正交试验确定了AB-8型大孔树脂纯化覆盆子总酚酸的最佳工艺条件:上样量为1.152 g·g^-1(生药量/树脂量),上柱液生药质量浓度为0.08 g·mL^-1,上柱液pH调为5-6,上柱吸附后静置30 min,上柱吸附速率为0.90-1.35 mL/min,解吸液为50%乙醇,解吸速率为0.45-0.90 mL/min,解吸液用量为5 BV,在上述最佳纯化条件下覆盆子总酚酸的吸附率为91.70%,解吸率为91.49%,覆盆子提取干浸膏中总酚酸含量由15.44%提高到54.18%。试验结果表明AB-8型大孔树脂可用于覆盆子总酚酸的纯化。The purification technology of total phenolic acid(TPA) in raspberry with macroporous resin was investigated.Using absorption and desorption ratio as evaluating indicators,the optimum purification conditions of TPA in raspberry with AB-8 type macroporous resin were obtained according to the single factor and orthogonal test.The loading quantity of sample(crude drugs weight/resin weight) was 1.152 g·g^-1 the concentration was 0.08 g·mL^-1,the pH of the sample was 5-6,static time after adsorption was 30 min,adsorption rate was 0.90-1.35 mL/min,concentration of ethanol as eluants was 50%,desorption rate was 0.45-0.90 mL/min and the volume of eluants was 5 BV.The absorption and desorption rate of TPA in raspberry was 91.70%and 91.49%under the optimum condition.The purity of TPA in dry extracts could be changed from 15.44%to54.18%,which indicated that AB-8 type macroporous resin could be applied for the purification of TPA in raspberry.
