灵芝孢子油抑制前列腺癌LNCaP细胞中AR激活及转录活性

摘　　要：目的:研究灵芝孢子油对双氰睾酮诱导的前列腺癌LNCaP增殖、雄激素受体AR的激活和转录活性及相关激酶的影响。方法:采用质粒转染、Western blot、CCK-8细胞增殖活性检测及双色荧光素酶报告基因活性等研究灵芝孢子油对雄激素依赖的前列腺癌LNCaP细胞的作用。结果:CCK-8检测显示灵芝孢子油明显降低DHT刺激的LNCaP细胞的增殖活性,同时,Western blot检测表明灵芝孢子油降低了DHT诱导的前列腺癌LNCaP细胞中AR和蛋白激酶D的磷酸化活性,而对其本底蛋白的表达水平无影响。荧光素酶报告基因活性检测进一步表明灵芝孢子油明显抑制DHT诱导的LNCaP细胞中AR的转录激活活性。结论:灵芝孢子油可能通过抑制AR的激活及其转录活性而抑制雄激素依赖的前列腺癌细胞的增殖。Objective：To investigate the role of ganoderma lucidum spore oil on tumor cell proliferation,androgen receptor activation and transcriptional activity as well as related kinase activation in prostate LNCa P cancer cells. Method： The effects of ganoderma spore oil on the LNCa P cells of the prostate cancer cells were studied by using plasmid transfection,Western blot,CCK-8 cell proliferation assay and dual luciferase reporter gene activity.Result：LNCa P cancer cells proliferation induced by DHT was significantly reduced by ganoderma lucidum spore oil. At the same time, the Western blot assay showed that ganoderma spore oil decreased the phosphorylation activity of AR and protein kinase D in prostate cancer LNCa P cells, but had no effect on the expression of the protein kinase DHT. Luciferase reporter gene activity assay indicated that the activity of AR in LNCa P cells induced by DHT was significantly inhibited by Ganoderma spore oil.Conclusion：Ganoderma lucidum spore oil may contributes to LNCa P cancer cells growth inhibition through down-regulation of AR activation and transcriptional activity in androgen dependent prostate cancer cells.
