摘 要:目的制作大鼠视神经横断损伤模型,在损伤视神经的同时保留视神经鞘,为视神经损伤后再生的研究提供模型基础。方法sD大鼠20只,随机分为3组:空白对照组2只,余18只右眼为实验组,左眼为对照组。将实验组与对照组分别按损伤后存活时间的不同分为7d组、14d组、30d组,每组6只。实验组在保留视神经鞘膜的前提下横断性切断视神经纤维;对照组单纯分离球后组织暴露视神经,不损伤视神经纤维。术后7d、14d、30d行光学相干断层扫描(OCT)检查;视网膜平铺荧光显微照相;视网膜组织病理切片。结果通过比较OCT检查;视网膜平铺RGC计数以及视网膜组织病理改变,可见术后不同时间点实验组大鼠的视网膜神经节细胞细胞排列紊乱、数目减少,视网膜变薄。而对照组与空白对照组未见明显的改变。结论于球后视神经2mm处切开视神经鞘膜,离断视神经纤维,创建了保留视神经鞘的视神经横断损伤模型,为临床进一步研究视神经损伤后的修复提供基础。Objective To provide a model for the research of optic nerve regeneration by setting up optic nerve transection injury model in rat and preserving the vaseulature and meninges. Methods Twenty healthy SD female rats were randomly divided into three groups: blank group (n = 2 ) , experimental group ( right eye, n = 18 ) , control group ( left eye, n = 18). According to survival time after the surgery, the rats in experimental group and control group were divided into 7 days, 14 days and 30 days groups with 6 rats in each group. The optic nerve fibers of rats in experimental group were completely transected, while the optic nerve sheath membranes were kept. The optic nerve of rats in control group were purely exposed. At one week, two weeks and three weeks after operation the optical coherence tomography ( OCT ), retinal flat fluorescent micrography and retinal tissue pathological slices were performed to observe the changes of the rats' retina. Results In experimental group the retinal thickness of the rats decreased, the RGC count gradually reduced and the arrangement of cells was disordered. There was no obvious changes in control group and blank group. Conclusion An optic nerve transection model in adult rat can be set up by opening the meninges of the optic nerve 2 mm behind the eyeball and keeping the vasculature and meninges. This model can provide a foundation for the research of optic nerve regeneration.
