摘 要:目的:准确理解氯丙嗪对体温调节功能的干扰作用,以及氯丙嗪的中枢神经系统不良反应,对实验中"氯丙嗪对中枢神经系统影响"的实验内容进行扩充及方法学改进。方法:在原有氯丙嗪对体温的调节实验中增设高温环境,同时增加小鼠木僵实验。选取48只雄性ICR小鼠,将其中32只随机分为生理盐水+低温环境组、0.075%氯丙嗪+低温环境组、生理盐水+高温环境组以及0.075%氯丙嗪+高温环境组,每组8只,检测正常体温后,给予氯丙嗪或等体积生理盐水,30 min后分别将小鼠放入人工低温或高温环境中,于10 min、20 min、30 min、40 min以及60 min检测小鼠体温;对其余16只小鼠进行木僵实验,给予小鼠0.4%氯丙嗪或生理盐水后,每隔10 min检测是否出现木僵状态。结果:注射0.075%氯丙嗪后,小鼠在人工高温环境中体温随时间延长逐渐升高,与正常环境组具有显著差异,而在人工低温环境中体温显著降低,呈现随环境温度的升降而升降的现象,充分体现了氯丙嗪对体温调节功能的干扰作用。注射0.4%高浓度氯丙嗪后20 min,小鼠出现持续的木僵状态。结论:经过改进后的实验方法能够较原有方法更能全面体现氯丙嗪对中枢神经系统的影响以及不良反应。Objective: To understand the comprehensive effect of chlorpromazine on body temperature regulating function and its central nervous system adverse effect, we added high temperature environment and the stupor experiment to students' experiment. Methods: Thirty four male ICR mice were randomly divided into normal saline+low environmental group; 0.075% chlorpromazine+low environmental group; normal saline+high temperature environment group; 0.075% chlorpromazine+high temperature environment group. After testing normal temperature, the mice were given chlorpromazine or equal volume of saline, 30 minutes later, they are transferred into the artificial low temperature or high temperature environment, then body temperature were tested at 10, 20, 30, 40, 60 minutes. 0.4% chlorpromazine or saline were given to other two groups of mice, then detect the stupor state every 10 minutes. Results: Injection of 0.075% chlorpromazine combined with high temperature environment caused a gradual rise in body temperature with the prolongation of time, and showed significantly difference to normal group, on the other hand, the mice injected of chlorpromazine combined with low environment temperature had significantly low temperature. The mice showed stupor state after 20 minutes after injection of 0.4% chlorpromazine. Conclusion: The current experiment can reveal the more comprehensive effects of chlorpromazine on central nervous system compare with the original experimental design.
