摘 要:试验以长白山野生白耙齿菌菌株为试验材料,进行ITS序列鉴定及人工驯化栽培试验。利用通用引物ITS1和ITS4进行ITS序列鉴定,同时利用农作物下脚料对白耙齿菌进行人工驯化栽培。原种培养料设置6个处理,栽培种培养料设置8个处理,通过ITS序列分析结果得出该菌株ITS序列与NCBI中登录号为JX290578.1、FJ462768.1、EU301643.1等9个白耙齿菌菌株相似性达100%,验证了其为白耙齿菌(Irpex lacteus);最佳原种培养料配方为木屑41.5%、豆秸41.5%、麦麸10%、黄豆粉5%、石膏1%、白糖1%,菌丝生长速度最快,为7.571 mm·d-1,且菌丝浓密,洁白,粗壮;最佳栽培种培养料配方为木屑54%、玉米芯29%、麦麸10%、黄豆粉5%、石膏1%、白糖1%,菌丝生长速度及长势显著高于其他处理。Taking Irpex lacteus strain of Changbai Mountain as the tested material, its molecular identification and artificial do- mesticated cultivation were studied. The ITS1 and ITS4 of universal primers were used to identify the sequence of ITS, at the same time, the agricultural residues were used in the I. lacteus domestication. We set 6 disposes for original media formula and 8 disposes for plant media formula, then each dispose have 5 times. The identities of ITS sequence indicate the tested material is 1. lacteus. It is similarity with nine I. lacteus of sequence, suce as JX290578.1, FJ462768.1, EU301643.1. The original media formula for I. lacteus cultivation is considered to be that within composed of sawdust 41.5%, beanstalk 41.5%, wheat bran 10%, soybean meal 5%, gypsum 1%, sugar 1%, and the mycelia had the fast growth speed with 7.571 mm·d^-1. The mycelia is very thick, white and strong. The most suitable medium is composed of sawdust 54%, corncob 29%, wheat bran 10%, soybean flour 5%, gypsum 1%, sugar 1%. Its mycelia growth rate was significantly higher than other disposes.
