正常结直肠组织、腺瘤、结直肠腺癌中miRNAs的表达差异

摘　　要：目的探讨6种microRNA(miRNA)在正常结直肠组织、管状腺瘤、管状绒毛状腺瘤及腺癌中的表达差异。方法共搜集44例管状腺瘤、16例管状绒毛状腺瘤、20例腺癌患者的病变组织及其旁正常组织标本,应用Real-time PCR方法分别检测miR-1、miR-9、miR-31、miR-135a、miR-137、miR-145共6种miRNA的表达量,分析表达差异。结果 1管状腺瘤中miR-1、miR-9、miR-137、miR-145的表达均低于瘤旁正常组织,miR-31、miR-135a的表达均高于瘤旁正常组织,其中miR-9、miR-135a差异有统计学意义(P<0.05),miR-1、miR-137、miR-145、miR-31差异无统计学意义(P>0.05)。2管状绒毛状腺瘤中miR-1、miR-137、miR-145的表达均低于瘤旁正常组织,miR-9、miR-135a、miR-31的表达均高于瘤旁正常组织,差异均有统计学意义(P<0.05)。3结直肠腺癌中miR-1、miR-9、miR-137、miR-145的表达均低于癌旁正常组织,miR-31、miR-135a的表达高于癌旁正常组织,差异均有统计学意义(P<0.05)。结论正常结直肠组织、结直肠腺瘤、结直肠癌中miRNA的差异表达,提示上述miRNA在从正常结直肠组织到结直肠腺瘤、结直肠腺瘤到结直肠癌的发生发展过程中可能发挥作用,为miRNA作为疾病预测、预后指标提供了实验基础。Objective To investigate the expression difference of microRNAs in colorectal tubular adenoma,tubulovillous adenoma and colorectal cancer.Methods Real time-polymerase chain reaction was used to analysis the expression level of miR-1,miR-9,miR-31,miR-135 a,miR-137 and miR-145 between colorectal tubular adenoma（n=44）,tubulovillous adenoma（n=16）,colorectal cancer（n=20）and paired paracancerous normal colorectal tissues.Results Compared with paired paracancerous normal colorectal tissues,colorectal tubular adenoma had decreased expression of miR-1,miR-9,miR-137 and miR-145 as well as increased expression of miR-31 and miR-135 a.Colorectal tubulovillous adenoma had decreased levels of miR-1,miR-137,miR-145,as well as increased levels of miR-9,miR-135 a,and miR-31.Colorectal cancer had the expressions of miR-1,miR-9,miR-137 and miR-145 decreased while the expression of miR-135 aand miR-31 increased.Conclusion The differential expressions of microRNAs suggested that miRNAs participate in the development of colorectal adenoma and cancer.
