摘 要:乳腺癌是女性肿瘤中发病率最高,且致死率仅次于肺癌的癌症种类。大部分乳腺癌病人死亡的主要原因是癌细胞向远端器官发生了转移。在本文中,我们研究了IL-13Rα2与乳腺癌转移以及病人不良预后的相关性,并初步探寻了其影响癌细胞转移的可能机制。通过实时荧光定量PCR、蛋白免疫印迹实验、Kaplan-Meier生存分析、细胞免疫荧光和细胞侵袭等实验方法,我们证明了IL-13Rα2的表达与ER阴性乳腺癌细胞的肺转移能力、ER阴性乳腺癌病人的肿瘤转移和不良预后正相关,与乳腺癌样本中ER的表达负相关,并且其中的作用机制可能与IL-13通过IL-13Rα2调节肿瘤细胞上皮-间质转换(EMT)有关。Breast cancer is the most frequent type of cancers in women in the world,and is the second cause of cancer deaths second only to lung cancer. The majority of patient deaths are due to the growth of disseminated tumor cells in distant organs. In this paper, we investigated the correlation of the expression of IL13Rα2 with breast cancer metastasis and poor prognosis, and preliminarily explored the mechanism of its effect on the metastasis of breast cancer cells. Through the methods such as re- al-time quantitative PCR, Western blot analysis, Kaplan-Meier survival analysis, immunofluorescence and matrigel invasion an- alyses, we demonstrated that the expression ofIL13Rα2 was positively correlated with the lung metastasis capacity of breast cancer cell lines as well as the metastasis and poor prognosis of ER negative breast cancer patients and negatively correlated with the ER expression in breast cancer patients. Furthermore, the mechanism was probably related to the regulation of epithe lial mesenchymal transition (EMT) by IL13 via IL13Rα2in cancer cells.
