Rho相关结构域BTB蛋白质1基因在胃癌组织中的表达及其临床意义

摘　　要：目的 探讨胃癌组织芯片中Rho相关结构域BTB蛋白质1（RhoBTB1）的表达与临床病理特征的相关性及其临床意义.方法 采用组织芯片技术建立67例胃癌患者的组织芯片,应用免疫组织化学技术检测芯片中RhoBTB1的表达.结果 RhoBTB1在胃癌组织中表达的阳性率低于癌旁黏膜组织[（53±29）％比（63±28）％,Z=2.59,P＜0.05];根据临床病理资料,胃癌组织中RhoBTB1的表达高低与病理分级明显相关,低/未分化腺癌RhoBTB1基因的阳性表达率低于中/高分化腺癌[（38±26）％比（65±25）％,Z=4.10,P＜0.05],但其表达与性别、TNM分期、浸润深度、淋巴结转移、脉管转移及远处转移等因素无明显相关（P＞0.05）;RhoBTB1的表达与年龄、淋巴结转移数目和肿瘤大小等无明显相关（P＞0.05）.结论 RhoBTB1可能是与胃癌发生相关的候选抑癌基因,检测该基因的表达有助于评估胃癌的生物学行为和预后.Objective To investigate the relationship between the expression of RhoBTB1 and the clinicopathological factors in gastric cancer tissue microarray and its clinical significance.Methods Tissue microarray technology was adopted to establish a tissue chip of 67 cases of patients diagnosed with gastric cancer and immunohistochemistry technique was used to detect the expression of RhoBTB1 in this chip.Results The expression level of RhoBTB1 in the gastric cancer tissue is lower than that in the nomal mucosa tissue adjacent to gastric cancer [（53 ± 29） % vs.（63 ± 28） %, Z =2.59, P 〈 0.05].The expression level of RhoBTB1 in the gastric cancer tissue might have correlation with pathological grades, besides, higher the grade, lower the expression level of RhoBTB1 [（38 ±26）% vs.（65 ±25）%, Z =4.10, P 〈 0.05].However, the expression level of RhoBTB1 has no correlation with gender, TNM staging, depth of invasion, lymphatic metastasis, vascular metastasis and distant metastasis, neither with continuous variables, such as age and tumor size （P 〉 0.05）.Conclusion RhoBTB1 might be one of candidate tumor suppressor genes that have correlation with the progression of gastric cancer.Detecting the expression level of RhoBTB1 might contribute to estimating the biological behaviors and prognosis of gastric cancer.
