摘 要:目的 采用蛋白质组学技术筛选并鉴定出膀胱移行上皮细胞癌的差异性表达蛋白.方法 选取膀胱移行上皮细胞癌患者血清40例(膀胱癌术前组),膀胱移行上皮细胞癌术后患者血清40例(膀胱癌术后组)及正常体检者血清20例(正常对照组).采用表面增强激光解吸电离飞行时间质谱(SELDI-TOF MS)技术筛选出差异性表达蛋白,并应用基质辅助激光解吸电离飞行时间质谱(MALDI-TOF/TOF MS)对筛选出的差异性表达蛋白进行鉴定.结果 对所得数据应用生物信息学方法进行分析,筛选出m/z位于5 735及10 510的蛋白/肽段在膀胱癌术前组(3 885.951 2±850.899 2、4271.2364±1 132.2998)高表达,而在膀胱癌术后组(1 553.981 0±720.517 9、2 961.454 5±633.8647)及正常对照组(944.494 8±209.345 7、2 512.183 1±166.043 4)相对低表达,三组数据两两比较,差异均有统计学意义(P<0.01).应用MALDI-TOF/TOF MS技术鉴定m/z位于5 735及10 510的蛋白/肽段分别为差异性表达蛋白为锌α2糖蛋白(ZAG)及载脂蛋白A1(ApoA1).结论 应用蛋白质组学技术筛选并鉴定出了膀胱癌患者血清中的差异性表达蛋白ZAG与ApoA1.Objective To screen and identify the differentially expressed proteins of bladder transitional cell carcinoma with proteomic techniques.Methods 100 cases were divided into three groups: the preoperative group (40 cases), the postoperative group (40 cases) and the control group (20 cases).Surface-enhanced laser desorption/ionization-time of flight (SELDI-TOF MS) technology was applied to detect the differentially expressed proteins.Finally, the differential spot was identified by matrix-assisted laser-desorptionionization time-of-flight / time-of-flight mass sctrometry (MALDI-TOF/TOF MS).Results Two differentially expressed protein peaks of m/z 5 735 and 10 510 were obtained with SELDI-TOF MS.There was a high expression in the preoperative group (3 885.951 2 ± 850.899 2 and 4 271.236 4 ± 1 132.299 8) , and there was a low expression in the postoperative group (1 553.981 0 ± 720.5179 and 2961.4545± 633.8647) and the control group (944.4948± 209.3457 and 2 512.183 1 ± 166.043 4).The results of each two groups had statistical differences (P 〈 0.01).And m/z 5 735 and 10 510 protein peaks were identified as zinc-alpha-2-glycoprotein (ZAG) and apolipoprotein A1 (ApoA1).Conclusion The ZAG and ApoA1 are screened and identified by proteomic techniques in bladder transitional cell carcinoma.
