摘  要:本文对毕赤酵母H工程菌30L发酵产植酸酶的发酵条件及该酶的热稳定性进行了研究。研究表明,在菌体湿重达到90g/L时进入补糖阶段,达到220g/L时补糖结束进入混饲阶段,有利于菌体的生长及产酶,诱导时最佳pH为4.7,植酸酶发酵活性达到25000U/g。对所产植酸酶进行热稳定性研究发现,80℃保温30min残留酶活在75%,在50%保温30min残留酶活在95%,具有良好的热稳定性。In this paper, the optimal fermentation conditions of recombinant pichia pastoris H producing heat - resistant phytase in 30L fermentation tank and thermal stability of the enzyme were studied. The results show that it's carbohydrate supplement stages when the strain wet weight is 90 g/L , and when the wet weight of strain is 220g/L, its carbohydrate supplement end into the mixed feeding stage. It is good for the strain growing and enzyme producing, the optimum pH for introduction is 4. 7, and the fermentation activity is 25000U/g. Research on heat - resistant phytase shows that phytase has good thermal stability, and its residual enzyme activity still has 75% at 80℃ (30 mins ) ,and 95% at 50℃ (30 rains ).
