线粒体丝氨酸蛋白酶在甲状腺乳头状癌中的表达水平及其临床意义

摘　　要：目的探讨线粒体丝氨酸蛋白酶(Omi/Htr A2)在甲状腺乳头状癌中的表达及其与临床病理的关系。方法采用免疫组织化学方法和蛋白印迹法检测120例甲状腺乳头状癌、40例结节性甲状腺肿及40例正常甲状腺组织中Omi/Htr A2的表达水平,并分析其与甲状腺乳头状癌的临床病理特征的关系。结果免疫组织化学结果表明,甲状腺乳头状癌组织中Omi/Htr A2蛋白阳性率(71.67%)较结节性甲状腺肿(30.00%)、正常甲状腺组织(32.50%)明显升高,差异有统计学意义(P<0.05)。蛋白印迹法检测结果表明,Omi/Htr A2蛋白在甲状腺乳头状癌组织中的表达量为(1.184±0.060),明显高于结节性甲状腺肿组织(0.599±0.096)和正常甲状腺组织(0.606±0.097),差异有统计学意义(P<0.05)。Omi/Htr A2蛋白的表达与甲状腺乳头状癌的包膜浸润、淋巴结转移、肿瘤临床分期及肿瘤分化有关(P<0.05)。结论 Omi/Htr A2蛋白的表达可能成为判断甲状腺乳头状癌恶化程度及预后的指标。【Objective】 To analyze the expression of mitochondrial serine protease（Omi / Htr A2） in papillary thyroid carcinoma and its correlation with clinicopathologic parameters.【Methods】 Immunohistochemistry and Western blot were used to assess the expression of Omi / Htr A2 protein in 120 cases of papillary thyroid carcinoma,40 cases of nodular goiter and 40 cases of normal thyroid tissue,and its associations with clinicopathologic parameters were analyzed.【Results】 Immunohistochemistry showed the expression of Omi / Htr A2 was positive in 71.67% of the papillary thyroid carcinoma tissue,which was significantly higher than that in the nodular goiter（30.00%） and nomal thyroid tissue（32.50%,P〈0.05）.Western blot showed the relative amount of Omi / Htr A2 protein in the papillary thyroid carcinoma tissue was（1.184 ± 0.060）,which was significantly higher than that in the nodular goiter（0.559 ± 0.096） and normal thyroid tissue [（0.606 ± 0.097）,P〈0.05].The expression of Omi / Htr A2 was correlated with extracapsular invasion,lymph node metastasis,clinical staging and tumor differentiation（P〈0.05）.【Conclusion】 The expression of Omi / Htr A2 protein may be a marker for judging the malignancy and prognosis of papillary thyroid carcinoma.
