摘  要:目的观察脑缺血后肿瘤坏死因子受体相关因子6(TRAF6)的表达变化,探讨木犀草素的脑保护机制研究。方法用线栓法制作大鼠大脑中动脉(MCAO)闭塞模型。采用Western blot观察TRAF6蛋白变化,比较各组脑梗死体积、患侧脑水肿和神经功能缺损评分。结果脑梗死后24h,TRAF6的蛋白表达明显上调(2.14±0.05);木犀草素可抑制脑梗死后TRAF6的表达(P<0.05);明显改善神经功能缺失、减轻脑水肿及减小脑梗死体积,(P<0.05)。结论木犀草素通过抑制TRAF6的表达起到了脑保护作用。Objective This study is to evaluate the changes of tumour-necrosis factor receptor-associated factor 6 ( TRAF6) after cerebral ischemia and to explore the underlying mechanisms of luteolin.Methods Male Sprague-Dawley rats were subjected to permanent middle cerebral occlusion ( MCAO ) .Luteolin was systemically administered to explore the effect on TRAF6 at 24 h after cerebral ischemia by western blot.The infarct volume, brain water content, and neurological deficits were measured.Results The expression of TRAF6 was up-regulated at 24h after cerebral ischemia (2.14±0.05).Luteolin down-regulated TRAF6 expression induced by cerebral ischemia (P〈0.05 ) .Meanwhile, luteolin dramatically alleviated neurological deficit, brain water content and infarct volume ( P〈0.05) .Conclusion Luteollin can protect the brain from the damage caused by ischemia, whose neuroprotection effect may be through downregulation of TRAF6.
