摘  要:目的探讨人端粒酶逆转录酶(human telomerase reverse enzyme,h TERT)调控Gli1的表达及其对胃癌细胞侵袭迁移的影响。方法采用p IRES2载体构建h TERT过表达质粒,qRT-PCR和Western blot检测h TERT对Gli1表达的影响;构建Gli1启动子荧光素酶质粒,检测h TERT对Gli1启动子荧光素酶活性的影响;构建Gli1干扰质粒,并与h TERT过表达质粒共转染SGC-7901细胞;Transwell实验和划痕实验检测胃癌细胞的侵袭、迁移能力的变化;结果过表达h TERT显著上调Gli1 mRNA和蛋白的表达(P<0.01),并显著增加Gli1启动子荧光素酶的活性(P<0.01);而si-Gli1显著抑制Gli1的表达;干扰Gli1的表达,减弱h TERT对胃癌细胞侵袭、迁移能力的影响。结论 h TERT通过促进Gli1的转录,增加Gli1的表达,进而增强胃癌细胞的侵袭迁移。Objective To investigate the mechanism by which human telomerase reverse enzyme (hTERT) regulates Glil expression to promote metastasis of gastric cancer cells. Methods After down- regulation of Glil expression with a specific siRNA against Glil, the migration and invasion of gastric cancer cells were detected by wound healing and Transwell assays. The protein expression of hTERT and Glil was detected by Western blotting. Ectopie expression of hTERT was performed using pIRES2 vector. After the dual lueiferase vector that contains the Glil promoter was eo-transfeeted into SGC-7901 cells, and the luciferase activity in the cells was measured. Results Si-Glil markedly inhibited the Glil expression, and the down- regulation of Glil significantly suppressed the migration and invasion of gastric cancer cells. Ectopic expression of hTERT significantly enhanced the expression of Glil at both mRNA and protein levels (P 〈 0. 01 ). Over-expression of hTERT also significantly enhanced the luciferase activity of Glil promoter ( P 〈 0.01 ). Conclusion hTERT may promote the migration and invasion of gastric cancer cells through enhancing Glil expression at transcriptional level.
