摘 要:目的探讨慢病毒介导碱性成纤维细胞生长因子(basic fibroblast growth factor,b FGF)基因修饰的骨髓基质细胞(bone marrow stromal cells,BMSCs)对脑梗死后动脉生成的影响。方法利用慢病毒载体介导b FGF基因修饰BMSCs,获得稳定转染b FGF的BMSCs。脑梗死后1 d立体定向移植至梗死灶周围。术后14 d后采用免疫组织化学染色法标记梗死灶周围微小动脉,并采用MCID计算机成像分析系统测量微小动脉的密度及直径。结果与对照组比较,BMSCs组及b FGF-BMSCs组微小动脉密度及直径明显增加(P<0.05),且b FGF-BMSCs组微小动脉密度及直径显著高于BMSCs组(P<0.05)。结论 b FGF修饰的BMSCs可有效促进脑梗死后动脉生成。Objective To investigate the influence of lentiviral vector mediated basic fibroblast growth factor( b FGF) gene modified bone marrow stromal cells( BMSCs) on arteriogenesis in rats with cerebral ischemia. Methods Rat BMSCs were lentivirally tranduced to express b FGF stably. They were transplanted into the ischemic boundary zone 1d after middle cerebral artery occlusion. The arteriole were labeled by immunohistochemistry staining14 days after MCAO and the arteriolar density and diameter were measured in the ischemic boundary( IBZ) using the MCID computer imaging analysis system. Results Compared with control group,treatment with BMSCs and b FGFBMSCs significantly increased the arteriolar density and diameter in the IBZ( P〈0. 05). The arteriolar density and diameter in b FGF-BMSCs group significantly increased than that in BMSCs group( P〈0. 05). Conclusions b FGF gene-modified BMSCs can efficiently improve arteriogenesis after stroke.
