摘  要:目的 探讨大黄素-8-0-β-D-葡萄糖甙(EG)对人卵巢癌细胞系SKOV3细胞凋亡及Bcl-2家族蛋白表达的作用.方法 取对数生长期的SKOV3细胞,分为对照组和实验组,实验组用不同浓度的EG分别处理24、48及72 h,采用噻唑蓝(MTT)法测定EG对SKOV3细胞增殖的影响;采用Hoechst染色观察SKOV3细胞的形态学变化;不同浓度EG处理SKOV3细胞48 h后,分别采用流式细胞仪检测细胞凋亡和周期;免疫印迹法检测半胱氨酸天冬氨酸蛋白酶caspase-3、-9,Bcl-2和(或)Bax蛋白表达情况.结果 EG(20、40及80 mg/L)能剂量和时间依赖性地降低SKOV3细胞的活力,20、40及80 mg/L的EG作用SKOV3细胞48 h后,细胞凋亡率显著增加,分别为23.8%、35.5%及59.6%,明显高于对照组(2.3%);实验组SKOV3细胞caspase的活性形式cleaved caspase表达量显著增加;促凋亡蛋白Bax表达量升高,而抗凋亡蛋白Bcl-2表达量降低,呈剂量依赖性.结论 EG能抑制SKOV3细胞的增殖并诱导其凋亡,其作用机制可能与增加细胞caspase活性、下调Bcl-2基因的表达及上调Bax基因有关.Objective To explore the effect of emodin-8-O-β-D-glucoside (EG) on cell apoptosis and expression levels of Bcl-2 family proteins in human ovarian cancer SKOV3 cells.Methods SKOV3 cells were cultured and divided into two groups (control group and experimental group).Cell viabilities were determined by the methyl thiazolyl tetrazolium (MTT) method;apoptosis and cell cycle were analyzed by flow cytometry;the changes of protein expression of cleaved caspase-3, cleaved caspase-9, Bcl-2 and Bax were detected by Western blot.Results From the data of MTT, the cell proliferation of human ovarian cancer SKOV3 cells was inhibited by EG (20, 40, 80 mg/L) in a dose-and time-dependent manner.Flow cytometry assays showed that EG significantly induced apoptosis in SKOV3 cells.48 h after treated with EG (20, 40, 80 mg/L) , the apoptosis rate of the experimental group were increased gradually, and they were 23.8% , 35.5% , 59.6%, respectively, which were higher than these of the control group significantly.The data of Western blot showed that EG down-regulated Bcl-2 and up-regulated cleaved caspase-3, cleaved caspase-9, Bax in a dose-dependent manner.Conclusions EG can inhibit the proliferation of SKOV3 cells and promote apoptosis, and the anticancer effect of EG may be associated with the down regulation of Bcl-2 expression and up regulation of Bax expression, as well as the increase of relative activity of caspase 3 and caspase 9.EG may be a promising antitumor agent for cancer treatment.
