摘　　要：目的探讨雷公藤红素(celastrol)对卵巢癌顺铂(cisplatin,DDP)耐药细胞SKOV3/DDP的抑制作用及其机制。方法 MTT法检测不同浓度雷公藤红素对SKOV3/DDP细胞增殖的影响;显微镜下观察不同浓度雷公藤红素对SKOV3/DDP细胞形态的影响;流式细胞术检测不同浓度雷公藤红素对SKOV3/DDP细胞周期及凋亡的影响;细胞划痕试验检测不同浓度雷公藤红素对SKOV3/DDP细胞迁移能力的影响;Transwell体外侵袭试验检测不同雷公藤红素对SKOV3/DDP细胞侵袭能力的影响;Western blot法检测SKOV3/DDP细胞中凋亡及侵袭相关蛋白的表达水平。结果经雷公藤红素作用后,SKOV3/DDP细胞的增殖抑制率显著上升(P<0.05);镜下观察SKOV3/DDP细胞数目显著减少(P<0.05);G1期细胞比例明显升高(P<0.05);细胞凋亡率显著增加(P<0.05);细胞迁移距离明显减小(P<0.05);穿膜细胞数目明显减少(P<0.05);SKOV3/DDP细胞中Cyclin A、p-AKT、NF-KB、MMP-2、MMP-9和P-GP蛋白的表达水平均明显下降(P<0.05)。结论雷公藤红素对SKOV3/DDP细胞抑制作用显著,为临床晚期失去手术机会卵巢癌患者的治疗提供了参考。Objective To investigate the inhibitory effect of celastrol on cisplatin-resistant ovarian cancer SKOV3 / DDP cells as well as the relevant mechanism.Methods The effect of celastrol at various concentrations on proliferation of SKOV3 / DDP cells was evaluated by MTT assay,while that on cell morphology by microscopy,that on cell cycle and apoptosis by flow cytometry,that on migration ability by cell streak test,and that on invasion ability by in vitro Transwell assay.The expression levels of proteins associated with apoptosis and invasion in SKOV3 / DDP cells were determined by Western blot.Results After treatment with celastrol,the inhibition rate of proliferation of SKOV3 / DDP cells increased significantly（P〈0.05）,while the cell count decreased significantly under microscope（P〈0.05）,the percentage of cells at G1 phase and cell apoptosis rate increased significantly（both P〈0.05）,and the migrated distance and transmembrane cell count decreased significantly（both P〈0.05）.However,the expression levels of cyclin A,p-AKT,NF-κB,MMP-2,MMP-9 and P-GP in SKOV3 / DDP cells decreased significantly（P〈0.05）.Conclusion Celastrol showed significantly inhibitory effect on SKOV3 / DDP cells,which provided a reference for the therapy of patients with ovarian cancer at late stage missing the chance of surgery.
