摘  要:目的利用细胞体外创伤模型探讨无机三氧化矿物聚合体(MTA)和BioAggregate(BA)对牙髓细胞增殖及覆盖创面能力的影响,并评价其生物相容性。方法在培养皿中形成牙髓细胞的融合培养物,去除宽3mm的细胞层条带,建立体外创伤模型。在培养皿中分别加入不同浓度的MTA和BA浸提液培养24h,分别为MTA组(MTA原液组、MTA1/2液组、MTA1/4液组,各9皿)和BA组(BA原液组、BA1/2液组、BA1/4液组,各9皿);以模型中加入含有10%FBS的培养液作为对照组(9皿):检测各组细胞增殖能力及迁徙覆盖能力。结果BA原液组较其他组细胞增殖能力增强(P<0.05);与对照组相比,MTA原液组、MTA1/2液组、BA原液组及BA1/2液组细胞迁徙覆盖能力均增强(均P<0.05);BA原液组细胞增殖能力及迁徙覆盖能力均优于MTA原液组(均P<0.05)。结论MTA和BA均具有良好的生物相容性,但BA要优于MTA。Objective To investigate the effects of mineral trioxide aggregate(MTA) and BioAggregate on the prolifer- ation and wound covering of human dental pulp cells (HDPCs) in an in vitro wound model, and evaluate its biocompatibility. Methods Wounds were induced by making a 3 mm incision in cell monolayers across the length of tissue culture plates. The wound monolayers were treated with various dilutions of MTA and BioAggregate extracts for 24h to assess the cell proliferation and wound covering. MTA groups (undiluted MTA group, 1/2 concentration MTA group, 1/4 concentration MTA group) and BA groups (undiluted BA group, 1/2 concentration BA group, 1/4 concentration BA group) were the experimental groups, and DMEM bovine serum(10%FBS) served as the control group. Results Only BA undiluted extract increased cell proliferation ability(P〈 0.05). Treatment with undiluted MTA extract, 1/2 concentration MTA extract, undiluted BA extract and 1/2 concentration BA extract all enhanced cellular wound covering of HDPCs compared with those in control group(P〈0.05). Cells exposed to undiluted BA extract displayed higher cellular ability of proliferation and wound filling compared with those exposed to undiluted MTA extract (P〈0.05). Conclusion MTA and BioAggregate both have favorable biocompatibility, but BioAggregate is better than MTA.
