摘  要:目的建立保健食品中牛磺酸的在线衍生高效液相色谱(HPLC)分析方法。方法使用超声法提取保健品中牛磺酸,样品提取液与牛磺酸标准液分别以邻苯二甲醛(OPA)为衍生剂利用液相色谱自动进样器的在线衍生功能进行柱前衍生后,采用agilent SB-C18(4.6 mm×250 mm,5μm)分离,以0.1 mo L/L磷酸二氢钾缓冲液(p H=6.0):四氢呋喃:甲醇=32:1:22为流动相,柱温30℃,流速1.0 m L/min,在330nm波长下检测衍生产物,以保留时间对牛磺酸进行定性,峰面积进行定量检测。结果方法学考察表明牛磺酸在0.012~0.480 mg/m L浓度范围内有良好线性关系(r=0.9999),RSD为0.69%~3.10%,加标回收率的范围在91.1%~109.3%,检出限为0.031 mg/100g。结论在线衍生高效液相色谱法样品前处理简单,且具有良好的精密度和准确度,避免了手动衍生所带来的精密度差的问题,取得满意的结果,适用于保健品中牛磺酸的测定。Objective To establish a method for determination of taurine in health foods by on- line derivation HPLC.Methods Ultrasound method was used for the extraction of taurine from heath foods, sample extractor and taurine standardsolution were derived with ortho-phthalaldehyde(OPA)by online derivation of auto sampler before column derivation, separatedby agilent SB- C18(4.6 mm × 250 mm, 5 μm). Using 0.1 mo L/L potassium dihydrogen phosphate buffersolution(p H=6.0):tetrahydrofuran: methanol=32:1:22 as the mobile phase, the oven temperature 30 ℃, flow rate 1.0 m L/min, and the detectivewavelength 330 nm, the external standard method was used. Results The methodological evaluation showed the linear range oftaurine was from 0.012 to 0.480 mg/m L(r =0.9999). The RSD was 0.69 %-3.10 %, recovery rate was 91.1 %-109.3 %, and thelimit of detection was 0.031 mg/100 g. Conclusion The method is simple, precise and accurate, and it could avoid the problemof poor precision by manually derived, it is suitable for content determination of taurine in heath foods.
