摘　　要：目的采用Meta分析方法评价TNF-α各位点基因多态性与开角型青光眼(OAG)易感性的关系。方法计算机检索Pubmed、Web of science和Embase英文数据库,筛选出有关TNF-α各位点多态性与OAG易感性的病例-对照研究,提取相关数据,以Stata 12.0软件合并分析比值比(OR)和95%置信区间(CI),并进行发表偏倚和敏感性分析。结果纳入TNF-α-308G/A、-857C/T、-863C/A和-238G/A位点多态性与OAG易感性Meta分析的文献分别有8篇、2篇、2篇、3篇;TNF-α4个单核苷酸位点各基因型与OAG发病风险无关;以种族和青光眼类型分层分析的结果显示,与G等位基因及GG基因型相比,携带A基因、AA+AG基因型的亚裔人群OAG发病风险分别增加1.75倍、2.02倍;携带相应基因型的个体,高眼压型青光眼(HTG)发病风险分别增加1.53倍、1.76倍。结论 TNF-α-308G/A基因多态性与HTG易感性以及亚裔人群OAG易感性有关。Objective To evaluate the association between the SNPs of TNF-α and the risk of open angle glaucoma （OAG） using Meta-analysis. Methods General searches of Pubmed, Web of science and Embase were performed to screen studies concerning relationships between SNPs of TNF-α and susceptibility of OAG After collecting related data, Stata 12.0 was applied to assess the pooled analysis of OR and 95% CI, publication bias and sensitivity. Results A total of 8, 2, 2 and 3 studies were pooled to evaluate the associations of 4 SNPs （-308G/A, -857C/T, -863C/A and -238G/A） and the risk of OAG, respectively; No significant association was observed between each SNP and the risk of OAG; If stratified by ethnicities and types of OAG, TNF-ct-308G/A was found to be associated with OAG, Compared with G allele and GG genotype, Asian individuals with A allele and AA＋AG genotype were 1.75, 2.02 times more susceptible to OAGmrespectively. In addition, individuals with A allele and AA＋AG genotype were 1.53, 1.76 times more susceptible to HTC, respectively. Conclusion The SNP of TNF-α-308G/A was significantly associated with the risk of HTG and Asian individuals＇ OAG .
