摘 要:目的观察胡桃醌对人卵巢癌SKOV3细胞增殖及细胞周期的影响。方法采用MTT法检测胡桃醌对卵巢癌SKOV3细胞生长抑制作用;采用流式细胞仪检测胡桃醌对卵巢癌SKOV3细胞周期的影响;采用Western Blot检测卵巢癌SKOV3细胞Cyclin D1和Cyclin E的表达。结果与对照组比较,不同剂量胡桃醌对卵巢癌SKOV3细胞具有抑制作用且呈剂量依赖性,IC50为30.13μmol/L,将细胞周期阻滞在G0/G1期。与对照组比较,胡桃醌处理组细胞周期蛋白Cyclin D1表达降低(P<0.05),而Cyclin E蛋白表达变化不显著。结论胡桃醌通过抑制Cyclin D1的表达抑制细胞周期G0/G1期阻滞,抑制细胞生长。Objective To observe the effect of Juglone on ovarian cancer SKOV3 cells proliferation and cell cycle. Methods The cell proliferation of SKOV3 was detected by MTF assay. The cell cycle examined by Flow cytometry. Cyclin D1 and Cyclin E protein expression were evaluated by Western Blot. Results Juglone could significantly inhibit the growth of SKOV3 cell in dose - depend- ent manner and the IC50 is 30.13μM. Flow cytometry detection showed that cell cycle was maintained at the G0/G1 phases. Com- pared with the control group, expression of Cyclin D1 protein was remarkably decreased ( P 〈 0.05 ) in Juglone - treated group but expression of Cyclin E was not notable. Conclusion Juglone can suppress the growth of ovarian cancer SKOV3 cell. Juglone can decrease the expression of Cyclin D1 and block the cell at G0/G1 phases, which might be the mechanism of Juglone effect.
