摘 要:目的:探讨非甾体类非特异性抗炎药吲哚美辛栓对急性胰腺炎大鼠、促抗炎因子的影响及其可能的机制。方法:48只雄性SD大鼠随机分为急性胰腺炎组,吲哚美辛治疗组和假手术组3组。各组于术后6 h、24 h两个时间点分批处死大鼠,检测血清淀粉酶(AMS)、血清TNF-α、IL-6、IL-4、IL-10水平,观察胰腺标本的大体改变并取胰腺组织经HE染色,采用改良Schmidt病理学评分。结果:吲哚美辛治疗组在血清淀粉酶、胰腺病理评分及促抗炎因子水平方面均较急性胰腺炎组明显改善,差异具有统计学意义。结论:吲哚美辛通过调节促炎和抗炎因子的水平,从而改善胰腺组织炎症反应程度,在调节免疫平衡方面起了一定的作用。Objective: To investigate the effects of indomethacin on the inflammatory and anti-inflammatory cytokines modulation in rats of acute pancreatitis and the potential mechanism of such nonsteroidal anti-inflammatory drugs. Methods: Forty-eight male SD rats were randomized into groups of acute pancreatitis( group AP),indomethacin therapy( group IN) and sham operation( group OP). Animals in the three groups were sacrificed respectively at 6 h and24 h after treatment for determination of serum-amylase( AMS) level. Enzyme-linked immunosorbent assay( ELISA) technique was used to detect the blood levels of TNF-α,IL-6,IL-4 and IL-10. Pathological changes in the pancreas were observed and examined by light microscopy together with modified Schmidt scoring. Results: Compared with the other two groups,indexes,including blood serum-amylase levels,pathological scores and the blood levels of TNF-α,IL-6,IL-4 and IL-10,were significantly improved in group IN,and the difference was statistically significant. Conclusion: Indometacin may improve the inflammation of pancreas through modulating the serum levels of both inflammatory and anti-inflammatory factors in rats with acute pancreatitis,which suggests that this nonsteroid can function in boosting the immune balance.
