Numb和α-catenin蛋白在胃癌中的表达及其临床病理意义

摘　　要：目的检测Numb和α-连环蛋白（α-catenin）在胃癌及其配对正常胃黏膜中的表达，并分析两者与胃癌临床病理因素的关系及其在胃癌发生发展中的作用。方法采用免疫组织化学方法检测109例胃癌标本和97例配对正常胃黏膜中Numb和α-catenin蛋白的表达，分析其与临床病理因素的相关性。结果胃癌组织中Numb和α-catenin的阳性表达率显著低于正常胃黏膜（60.6%∶100.0%，χ2=48.361，P=0.000；76.1%∶100.0%，χ2=26.480，P=0.000）。Numb蛋白的表达与Lauren分型（χ2=17.018，P=0.000）、管状腺癌分化程度（χ2=17.586，P=0.000）相关，α-catenin蛋白的表达与Borrmann分型（χ2=6.700，P=0.035）、Lauren分型（χ2=11.098，P=0.001）、管状腺癌分化程度（χ2=8.203，P=0.017）、淋巴结转移（χ2=6.402，P=0.011）相关。109例胃癌组织中Numb与α-catenin表达呈正相关（rk=0.184，P=0.028）。结论胃癌组织Numb和α-catenin的表达均显著下调，且两者表达呈正相关，提示两者可能参与调控胃癌的发生发展，Numb蛋白可能通过α-catenin调节途径参与Wnt-β-catenin通路，进而在促进胃癌细胞增殖及侵袭转移中发挥重要作用。ObjectiveTo detect the expressions of Numb and α-catnin in the gastric cancer and matched normal gastric mucosa, and to analyze their relationship with clinicopathological factors of gastric cancer and explore their roles in gastric carcinogenesis and progression. MethodsImmunohistochemical method was used to detect the expressions of Numb and α-catnin in 109 cases of tumor specimens and 97 cases of matched normal gastric mucosa. The correlations between Numb, α-catnin and clinicopathological factors were analyzed. ResultsThe positive rates of Numb and α-catnin in gastric cancer were significantly lower than those in normal gastric mucosa （60.6%∶100.0%, χ2=48.361, P=0.000; 76.1%∶100.0%, χ2= 26.480, P=0.000）. The expression of Numb protein was correlated with Lauren type （χ2=17.018, P=0.000） and tubular adenocarcinoma differentiation （χ2=17.586, P=0.000）. The expression of α-catnin protein was correlated with Borrmann type （χ2=6.700, P=0.035）, Lauren type （χ2=11.098, P=0.001）, tubular adenocarcinoma differentiation （χ2=8.203, P=0.017） and lymph node metastasis （χ2=6.402, P=0.011）. The expressions of Numb and α-catnin were statistically correlated in 109 cases of gastric cancer （rk=0.184, P=0.028）. ConclusionCompared with normal gastric mucosa, both Numb and α-catnin expressions are downregulated and the two expressions are correlated in gastric cancer. Numb and α-catnin may be involved in the regulation of histological differentiation of gastric cancer by certain passages, Numb protein may be adjusted by α-catenin pathway which is involved in Wnt-βcatenin pathway, and thus plays an important role in promoting cell proliferation, invasion and metastasis in human gastric cancer.
