摘 要:目的:研究鼠尾草酚对卵巢癌生长的影响及其机制。方法:不同浓度的鼠尾草酚(2.5μmol·L^(-1)、5μmol·L^(-1)、10μmol·L^(-1))作用卵巢癌细胞株SKOV3 24 h后,CCK-8法检测细胞增殖;流式细胞术检测细胞凋亡;Western blot检测SKOV3细胞中PI3K、P-Akt、FOXO3a(Forkhead box O3,FOXO3a)、X连锁凋亡抑制蛋白(X-linked inhibitor of apoptosis protein,XIAP)、Akt蛋白的表达。建立起裸鼠卵巢癌皮下移植瘤模型,观察不同浓度的鼠尾草酚对卵巢癌皮下移植瘤生长的影响;免疫组织化学法检测肿瘤组织中FOXO3a、XIAP阳性表达。结果:与对照组相比,鼠尾草酚可明显抑制卵巢癌SKOV3细胞生长,并显著增加癌细胞凋亡率,呈浓度依赖性;鼠尾草酚可下调PI3K、P-Akt、XIAP蛋白表达,同时上调FOXO3a。鼠尾草酚各剂量组卵巢癌组织中XIAP阳性表达率均低于对照组,而FOXO3a阳性表达率均高于对照组。结论:鼠尾草酚可抑制卵巢癌细胞的生长,其机制可能通过PI3K/Akt信号通路,诱导其凋亡。Objective: To explore the effect and the mechanism of Carnosol on growth inhibition of ovarian cancer.Methods: After ovarian cancer cells SKOV3 were treated with different concentrations of Carnosol( 2. 5 μmol · L^(-1),5μmol·L^(-1),10 μmol·L^(-1)) for 24 h,cell growth was detected by the CCK-8 assay and apoptosis was evaluated by the flow cytometry. The expressions of PI3 K protein,P-Akt protein,Akt protein,FOXO3 a protein and XIAP protein in SKOV3 cells were measured by Western blot assay. SKOV3 cells were injected into nude mice to establish engrafted ovarian tumor model. Tumor weight of the mice was evaluated after being sacrificed. Immunohistochemistry was used to detect the positive expressions of FOXO3 a and XIAP in the ovarian tumors. Results: Compared with control group,Carnosol obviously suppressed the proliferation of SKOV3 cells and increased the apoptosis in a concentration-dependent? way. Carnosol can lessen the expressions of PI3 K protein,P-Akt protein and XIAP protein while increase the expression of FOXO3 a.All groups treated with Carnosol increased FOXO3 a expression and decreased XIAP in ovarian tumors as well. Conclusion: Carnosol can suppresse the proliferation of ovarian cancer cells by inhibiting the expressions of PI3 K and P-Akt protein to promote apoptosis.
