宫颈癌组织中D2-40标记淋巴管密度与高迁移率族蛋白B1表达的关系研究

摘  要:目的探讨宫颈癌组织中淋巴管密度(LVD)与高迁移率族蛋白B1(HMGB1)表达的关系。方法采用免疫组化法检测86例宫颈癌患者正常组织与癌组织中的D2-40(LVD特异性标记物)表达情况,计算组织LVD;用Western blot法检测正常组织和癌组织中HMGB1表达情况,并进行定量分析;用Pearson相关分析法分析LVD与HMGB1之间的相关性。结果宫颈癌患者癌组织中LVD及HMGB1表达水平均显著高于正常组织(P均<0.05),Ⅲ+Ⅳ期患者癌组织中LVD及HMGB1表达水平均显著高于Ⅰ+Ⅱ期患者(P均<0.05);癌组织中LVD与HMGB1表达水呈显著正相关关系(r=0.97,P<0.05)。结论宫颈癌组织中LVD与HMGB1的表达呈显著正相关关系,与肿瘤的发展密切相关,二者可能通过相互作用,共同促进肿瘤细胞的侵袭、转移等活动。Objective It is to investigate the relationship between the expression of lymphatic vessel density( lymphatic vessel density,LVD) and high mobility group protein B1( High mobility group protein box-1,HMGB1) in cervical cancer.Methods The expression of D2-40( LVD specific markers) in normal tissues and tumor tissues of patients with cervical cancer were detected by immunohistochemical method,LVD expression of HMGB1 in normal tissues and tumor tissues was detected by Blot Western,quantitative analysis; Correlation analysis between HMGB1 and LVD was analyzed by pearson correlation.Results The levels of LVD and HMGB1 in tumor tissues of patients with cervical cancer were significantly higher than that in normal tissues( P all 〈0. 05),at the same time,the levels of LVD and HMGB1 in tumor tissues of patients with Ⅲ and Ⅳstage was significantly higher than that in ones with stage Ⅰ and Ⅱ( P all 〈0. 05); The expression level of LVD in cervical cancer tissues was positively correlated with the relative expression level of HMGB1 protein( r = 0. 97,P 〈0. 05). Conclusion The correlation of expression of LVD and HMGB1 was significantly positive in cervical cancer,and the expression levels was closely correlated with tumor stage,they can work together to promote the invasion and metastasis of tumor cells.
