摘  要:目的研究ECDI处理的脾细胞对同种异体皮片存活的影响,并初步探讨其机制。方法建立小鼠尾部全厚皮片移植模型,随机法分5组。control组:免疫排斥组;rapa组:单独使用3mg/kg雷帕霉素组;ECDI.SPstwice组:移植前7d及移植后1d输注10^8 ECDI—SPs联合雷帕霉素组;SPs组:移植前7d及移植后1d输注未经处理脾细胞联合雷帕霉素组;ECDI—SPsx3组:移植前7d、移植后1d及7d3次输注ECDI—SPs联合雷帕霉素组。结果ECDI处理的脾细胞迅速发生了凋亡,ECDI—SPs两次输注明显延长了同种异体小鼠皮片存活时间(P〈0.05),ECDI—SPs3次输注皮片存活同2次输注差异无统计学意义(P〉0.05)。ECDI-SP输注后小鼠体内调节性T细胞比例增加(P〈0.05)。结论ECDI-SPs联合雷帕霉素能延长同种异体小鼠皮片存活时间,可能机制与调节性T细胞升高有关。Objective To investigate the effect of ECDI-SPs on the survival of a full-thickness skin allograft and its mechanism. Methods Full-thickness skin transplantation was established and mice were divided randomly into five groups; Control group: the skin allograft without treatment; Rapa group: skin allograft with 3 mg/kg rapamycin for 28 days ; ECDI-SPs twice group : ECDI-treated splenocytes were injected i.v. on day 7 before ( - 7) and 1 day after ( + 1 ) skin transplantation with 3 mg/kg rapamycin for 28 days ; SPs group : Untreated splenocytes were injected the same way and time as ECDI-SPs twice group with 3 mg/kg rapamycin for 28 days; ECDI-SPs x3 group: Another time of ECDI-treated splenocytes were injected 7 day after skin transplantation on the basis of ECDI-SPs twice group. Results Infusions of donor splenocytes treated with ECDI prolonged skin allograft survival significantly ( P 〈 0.05 ), and there were no significant difference between ECDI-SPs twice and x3 group ( P 〉 0.05), the proportion of regulatory T cells increased in ECDI-SPs twice and x3 group ( P 〈 0.05). Conclusion Pre-and post-transplant infusions of splenocytes treated with ECDI prolonged skin allograft survival probably by up-regulation of regulatory T cells in spleen.
