摘 要:目的研究miR-429在肾透明细胞癌中的表达,及其对肾透明细胞癌Caki-1细胞侵袭能力的影响。方法实时定量PCR检测肾透明细胞癌组织中miR-429基因的表达水平。培养肾透明细胞癌Caki-1细胞,将mi R-429的前体转染Caki-1细胞,检测转染后Caki-1细胞的侵袭能力。结果与癌旁组织相比,miR-429在肾透明细胞癌组织中的表达下调显著,且与肾透明细胞癌的分级和淋巴结转移相关。miR-429的前体能够使Caki-1细胞穿透滤膜的数量明显减少。结论 miR-429能够抑制肾透明细胞癌细胞侵袭,发挥转移抑制基因的作用。Objective To explore the expression of miR-429 in renal clear cell carcinoma(Caki-1), and the effect of miR-429 on the invasive ability of Caki-1 cells. Methods Real-time PCR was used to detect the expression of miR-429 in renal clear cell carcinoma and the adjacent tissues. The Caki-1 cells were cultured, and transfected with miR-429 precursor, and then the invasive ability of Caki-1 cells were detected by transwell assay. Results The expression level of miR-429 was down-regulated in Caki-1 cells than in the normal adjacent tissues, with a negative relationship with the grades and metastasis of lymph node. The miR-429 precursor could decrease the number of trans-membrance cells in Trans-well assay. Conclusion miR-429 inhibits the invasive ability of Caki-1 cells, plays a role as metastasis inhibitive gene in renal clear cell carcinoma cells.
