摘  要:采用3种不同浓度抗氧化剂维生素C(VC)、聚乙烯吡咯烷酮(PVP)、硫代硫酸钠(Na2S2O3)处理褐化的耐盐柳树海柳1号愈伤组织,检测结果表明:在PVP浓度为8mg·L-1时,总酚含量下降显著,与对照相比下降了86.8%;与愈伤组织褐化相关的超氧化物歧化酶(SOD)和过氧化氢酶(CAT),在PVP为8mg·L-1时活性分别达到最高,与对照相比升高了94.8%和79.7%;且多酚氧化酶(PPO)和过氧化物酶(POD)的活性,在PVP为8mg·L-1处理时,活性达到最低,与对照相比降低了59.8%和67.6%。相关性分析结果表明:愈伤组织褐化程度与总酚含量、POD和PPO活性呈显著正相关,而与SOD、CAT活性则呈负相关。说明PVP处理海柳1号愈伤组织,能有效抑制总酚的积累,降低其褐化程度。Different concentrations of vitamin C(VC),polyvinylpyrrolidone(PVP)and sodium thiosulfate(Na2S2O3)were used to treat browning calli of‘Salix Hailiu 1'.Results showed that the total phenols content were the lowest and decreased by 86.8% at 8 mg·L^-1 PVP.The activities of superoxide dismutase(SOD)and catalase(CAT)were the highest and increased by 94.8%and 79.7%compared with CK at 8mg·L^-1 PVP.The The activities of polyphenol oxidase(PPO)and peroxidase(POD)were the lowest and decreased by 59.8%and 67.6%compared with CK at 8mg·L^-1PVP.Correlation analysis results showed that the levels of callus browning were positively correlated with the total phenols content and the activities of POD and PPO,but negatively correlated with the activities of SOD and CAT.The levels of callus browning of Salix Hailiu 1could be inhibited by the addition of PVP in the medium.
