摘 要:目的观察在高压缺氧条件下视网膜Müller细胞谷氨酸(Glu)摄取功能改变,以及通窍活血方对其影响。方法体外纯化培养7天SD大鼠视网膜Müller细胞至P2代;采用中药血清药理学方法制备通窍活血方含药血清;根据[3H]标记的D,L-Glu摄入量判断Müller细胞谷氨酸(Glu)摄入功能。结果高压缺氧条件下Müller细胞谷氨酸(Glu)摄取功能明显下降(P<0.05);通窍活血方含药血清可明显改善高压缺氧条件下Müller细胞谷氨酸(Glu)摄取功能(P<0.05)。结论通窍活血方含药血清可明显改善高压缺氧条件下Müller细胞谷氨酸(Glu)摄取功能,这可能是通窍活血复方对青光眼患者视神经保护的药物干预途径之一。Objective: To observe the effect of Tongqiao Huoxue Decoction on the altered glutamate intakes under the conditions of high pressure and hypoxia. Methods: SD rats and P2 generation of retinal Mailer cells were produced by vitro purification culture of seven days; the medicinal serums of dredging orifices and activating blood circulation were prepared by serum pharmacology of Chinese medicine ; the intake amount of L-Glu can be used to judge the absorbing function of Mailer cells according to the labeled D by [3HI. Results: The absorbing function of glutamate (Glu) in Mailer cells under the conditions of high pressure and hypoxia significantly decreased ( P〈0.05 ) ; Tongqiao Huoxue Decoction containing serum can significantly improve the absorbing function of glutamate (Glu) in Mailer ceils under the conditions of high pressure and hypoxia ( P〈0.05 ). Conclusion: Tongqiao Huoxue Decoction containing serum can significantly im- prove the absorbing function of glutamate (Glu) and it may be one of the herbal interventions for patients with glaucoma optic neuroprotective drug.
