摘 要:目的:研究黄芪多糖(Astragalus polysaccharide,APS)对胰岛素抵抗H9c2(IR-H9c2)心肌细胞葡萄糖摄取的影响,并探讨其信号机制。方法:采用高浓度胰岛素处理复制IR-H9c2心肌细胞模型,给予一定剂量的APS处理,MTT法筛选合适的实验浓度,2-脱氧-[3H]-D-葡萄糖法检测细胞的葡萄糖摄取,Western blotting法检测腺苷酸活化蛋白激酶(AMP-activated protein kinase,AMPK)的磷酸化水平。结果:APS可以明显促进IR-H9c2心肌细胞对葡萄糖的摄取,并能显著提高其AMPK的活性,且AMPK抑制剂Compound C明显降低了APS刺激IR-H9c2心肌细胞葡萄糖摄取的作用。结论:APS可以促进IR-H9c2心肌细胞的葡萄糖摄取,这一作用与AMPK的活化有关。Objective To explore the effect of astragalus polysaccharide (APS) on glucose uptake of insulin-resistant H9c2 cardiomyocytes (IR-Hgc2) and its possible signal mechanism in vitro. Methods IR-Hgc2 models were induced by high concentration insulin. The models were cultured in a media containing APS. The activity of IR-H9c2 was detected by MTT. The glucose uptake was measured by the 2-deoxy-[3H]-D-glucose method. The phosphorylation of AMP-activated protein kinase (AMPK) was examined by Western blotting. Results APS could significantly increase the glucose uptake and AMPK activity of IR-Hgc2. The APS-stimulated glucose uptake was significantly attenuated by pretreatment with Compound C, a selective AMPK inhibitor. Conclusion APS could stimulate the glucose uptake in IR-H9c2 ,which may correlate with the activation of AMPK.
