摘 要:目的观察金属蛋白酶组织抑制物-1(TIMP-1)以及细胞外信号调节蛋白激酶-1(ERK-1)在视网膜光损伤中的表达情况,分析视网膜光损伤中促红细胞生成素(EPO)对TIMP-1以及ERK-1表达的影响。方法选取健康成年大鼠60只作为实验动物,随机选取10只作为空白对照组(不进行任何干预),其余50只大鼠建立视网膜光损伤模型后,随机分为模型对照组(给予光照)和观察组(给予光照结合注射EPO)共两组,每组25只。在光照后的12 h、2d、3 d、4 d以及7 d时随机处死5只大鼠,应用HE染色处理后观察视网膜色素上皮细胞细胞(RPE)形态学变化,同时应用免疫组织化学法测定TIMP-1以及ERK-1表达情况。结果与空白对照组相比,视网膜光损伤大鼠的视网膜RPE细胞结构遭受明显破坏,并且随着时间的延长,损伤逐渐加重,但是观察组大鼠的细胞破坏程度显著轻于模型对照组;空白对照组无TIMP-1以及ERK-1阳性表达,模型对照组仅见少量TIMP-1以及ERK-1阳性表达,与空白对照组相比,无显著性差异(P>0.05),在相同时间点上,观察组大鼠的TIMP-1以及ERK-1显著高于其他两组(P<0.01),直线相关分析显示观察组大鼠的TIMP-1阳性表达水平与ERK-1阳性表达水平呈显著正相关(P<0.05)。结论视网膜光损伤能够导致视网膜RPE遭受严重破坏,给予EPO干预能够显著提高TIMP-1以及ERK-1阳性表达水平,有效改善视网膜光损伤程度,保护视网膜RPE细胞。Objective To observe TIMP - 1 ( tissue inhibitor of metalloproteinase - 1 ) and ERK - 1 ( extracellular signal - regulated pro- tein kinase - 1 ) expressions in retinal photic injury and analyze the effect of EPO (Erythropoietin) on the expressions of TIMP - 1 and ERK - 1. Methods 60 healthy adult rats were selected as experiment animal. 10 ruts were randomly selected as blank control group (without any interven- tion), the other 50 rats were established as retinal photic injury model. They were randomly divided into two groups, model control group (gave the light) and observation group (giving light and EPO injection). There were 25 rats in each group, 5 rats were random executed after illumina- tion in 12 h, 2 d, 3 d, 4 d and 7 d. After the application of HE staining processing, the retinal pigment epithelial cell (RPE) morphology changes were observed, and TIMP - 1 and ERK - 1 expressions were determined by immunohistochemical method. Results When compared with the blank control group, RPE ceils structure in rats was destroyed significantly in retinal photic injury. With the extension of time, the damage aggra- vated gradually, but the observation group rats were significantly lighter than the destruction of cells in model control group. There were no TIMP -1 and ERK- 1 positive expressions in blank control group, there were only a small amount of TIMP - 1 and ERK - 1 positive expressions in model control group, when compared with the blank control group. There was no significant difference ( P 〉 0.05 ). At the same time point, TIMP - 1 and ERK - 1 of observation group were significantly higher than that of the other two groups ( P 〈 0.01 ), linear correlation analysis showed that the TIMP - 1 expression level and ERK - 1 expression level was positively correlation in observation group ( P 〈 0.05 ).
