摘 要:目的研究RAD51基因G135C多态性和XRCC3基因T241M多态性与卵巢癌的相关性。方法对70例卵巢癌和70例健康对照人群的样本分别进行DNA提取、RAD51基因和XRCC3基因扩增及测序,再结合人群临床资料进行统计学分析。结果 RAD51基因型G/G、G/C和C/C在病例组的分布频率分别为71.4%、20.0%和8.6%,在对照组的分布频率分别为91.4%、8.6%和0。病例组突变型频率显著高于对照组(<0.05)。XRCC3基因T/T、T/M和M/M在病例组的分布频率分别为88.6%、8.6%和2.9%,在对照组的分布频率分别为85.7%、14.3%和0;两组差异无统计学意义(>0.05)。RAD51基因型G/G在卵巢浆液性癌组和黏液性癌组中的分布频率分别为68.0%和80.0%,变异基因型(G/C^+C/C)分别为32.0%和20.0%。RAD51 G135C基因型在不同的卵巢癌组织学类型间的分布频率差异无统计学意义(>0.05)。XRCC3基因T/T在卵巢浆液性癌组和黏液性癌组中的频率分别为92.0%和80.0%,变异基因型(T/M和M/M)分别为8.0%和20.0%。XRCC3基因T241M基因型在不同的卵巢癌组织学类型间的分布频率差异无统计学意义(>0.05)。结论 RAD51基因G135C多态性与卵巢癌发病有相关性,XRCC3基因T241M多态性与卵巢癌易感性无显著相关性。两基因的野生型及突变型与卵巢癌的组织学类型不相关。Objective To study the correlation between G135C RAD51 gene polymorphism and T241M XRCC3 gene polymorphism and ovarian cancer. Methods Seventy cases of ovarian cancer and 70 cases of healthy controls were selected for DNA extraction respectively; and RAD51 gene and XRCC3 gene were amplified and sequenced for statistical analysis combined with clinical data. Results Distribution frequency ofRADS1 genotype G/G, G/C and C/C in case group were 71.4%, 20.0% and 71.4% respectively, and the distribution frequency in the control group were 91.4%, 8.6% and 0; mutant frequency in case group was significantly higher than that of control group (P 〈 0.05). Distribution frequency of XRCC3 gene T/T, T/M and M/M in the case group were 88.6%, 8.6% and 2.9%, and the distribution of frequency in the control group were 85.7%, 14.3% and 0; there were no statistical significance (P 〉 0.05). Distribution ofRAD51 G/G genotype in ovarian serous carcinoma and mucous carcinoma group were 68.0% and 80.0% respectively, and the distribution frequency of mutation genotype (G/C+C/C) were 32.0% and 20.0% respectively; there were no statistical significance of RAD51 G135C ganotype among different ovarian cancer histology (P 〉 0.05). Distribution ofXRCC3 gene T/T in ovarian serous carcinoma group and the frequency of mucous carcinoma group were 92.0% and 80.0% respectively, and the mutation genotype (T/M and M/M) were 8.0% and 20.0% respectively; there were no statistical significance ofXRCC3 gene T241M type among different ovarian cancer histology (P 〉 0.05). Conclusions G135C RADS1 gene polymorphisms are associated with ovarian cancer, and there are no significant correlation between XRCC3 gene polymorphisms and ovarian cancer susceptibility. There are no associations between wild type and mutant of RAD51 gene and XRCC3 gene and histological types of ovarian cancer.
