葡萄糖调节蛋白78和钙网蛋白在喉鳞状细胞癌组织中的表达及意义

摘　　要：目的探讨葡萄糖调节蛋白78(GRP78)及钙网蛋白(CRT)与喉鳞状细胞癌之间的关系,从而为喉凝状细胞癌的临床诊治及治疗提供新思路。方法采用免疫组化方法及Western blot方法检测60例喉癌患者喉癌组织及癌旁组织中的GRP78与CRT的表达,探讨GRP78与CRT与喉癌病理特征之间的关系。结果 CRP78和CRT在喉癌组织中阳性细胞平均光密度值分别为0.3585±0.0006和0.2962±0.0126,在喉癌癌旁组织中分别为0.331±0.006和0.2847±0.0043,CRP78和CRT在喉癌中的平均光密度值高于癌旁组织,差异均具有统计学意义(P<0.005)。采用Pearson相关分析统计分析数据结果显示:CRP78及CRT与喉鳞状细胞癌密切相关,CRP78在喉凝状细胞癌中的表达随CRT升高而增加(r=1.45,P<0.001)。CRP78及CRT在喉癌组织中的平均灰度值分别为0.53±0.21和0.65±0.23,在正常组织中分别为0.24±0.14和0.56±0.17,CRP78及CRT在喉癌组织中的平均灰度值显著高于癌旁组织,差异均有统计学意义(P<0.05)。结论 CRP78及CRT在喉癌中呈高表达,与喉癌的发生发展密切相关,且两者在喉凝状细胞癌组织中的表达量呈正相关。Objective To investigate the relationship between the biological behavior of GRP78 and CRT in squamous cell carcinoma of larynx,which provides new ideas and approaches for the diagnosis and treatment of laryngeal carcinoma. Methods The expression of GRP78 and CRT by immunohistochemical method and Western blot method for laryngeal cancer tissue of patients with laryngeal carcinoma and 60 cases of detection of adjacent tissues,and to investigate the relationship between GRP78 and CRT and laryngeal pathological features. Results The average optical density value of CRP78 and CRT in laryngeal carcinoma positive cell were 0. 3585 ± 0. 0006 and 0. 2962 ± 0. 0126,were 0. 331 ± 0. 006 and 0. 2847 ± 0. 0043 in laryngeal carcinoma paracancerous tissue. The differences were statistically significant in expression of CRP78 and CRT in laryngeal carcinoma（ P 〈 0. 005）. Pearson correlation analysis showed that： CRP78 and CRT have relationship with laryngeal squamous cell carcinomas with their level of expression. The laryngeal squamous cell carcinoma pathological classification were positively correlated（ r = 1. 45,P 〈0. 001）. CRP78 and CRT in laryngeal carcinoma average gray values were 0. 53 ± 0. 21 and 0. 65 ± 0. 23,respectively 0. 24 ± 0. 14 and 0. 56 ±0. 17 in normal tissue. The differences were statistically significant in expression in laryngeal carcinoma,the（ P 〈 0. 05）. Conclusion CRP78 and CRT expression in laryngeal carcinoma is closely related to the occurrence and development of laryngeal carcinoma,and the expression of the two condensate in the throat- cell carcinoma rare positively correlated.
