摘 要:目的:探讨血管性痴呆(VD)模型大鼠海马CA1区P精蛋白(P-GP)及谷氨酸转运体1(GLT-1)长时间表达变化及其神经元保护机制。方法:采用反复脑缺血再灌注复制VD大鼠模型在15 d和1月两个时间点采用Morris水迷宫检测大鼠空间学习记忆能力,用神经颗粒素(Ng)/P-GP和GFAP/GLT-1免疫荧光双标法,观察海马区神经元P-GP和胶质细胞GLT-1的表达变化。结果:(1)模型组大鼠各时间点的Morris水迷宫逃逸潜伏期比假手术组均明显延长(P<0.01)。(2)与假手术组相比,模型组大鼠海马CA1区Ng/P-GP阳性神经元的数量明显降低(P<0.01)而GFAP/GLT-1阳性胶质细胞数以及海马区神经元上P-GP和胶质细胞上GLT-1表达均明显升高(P<0.01);与模型组15 d时间点比较,模型组1月时间点的海马CA1区Ng/P-GP阳性神经元的数量明显降低而GFAP/GLT-1阳性胶质细胞数以及P-GP和GLT-1的表达均明显升高(P<0.01)。(3)积分光密度值测定显示:模型15 d时间点Ng/P-GP和GFAP/GLT-1阳性细胞的IOD值比假手术组各时间点均明显变大(P<0.01);模型1月时间点Ng/P-GP和GFAP/GLT-1阳性细胞的IOD值较15 d时间点更大(P<0.01)。结论:神经元的P-GP和胶质细胞的GLT-1可能参与VD大鼠海马神经元的保护。Objective:To observe the dynamic changes of P-glycoprotein(P-GP) and glutamate transporter 1(GLT-l)expression in hippocampal CA1 region in vascular dementia(VD) rats at a long time points and the neuronal protection mechanism therein.Methods:VD models were established by repeated cerebral ischemia-reperfusion.At 15 d and 1month,Morris water maze was used to detect the spatial learning and memory ability,double immunofluorescence for neurogranin/P-GP and GFAP/GLT-l was performed to observe the changes of neuronal P-GP and glial GLT-l expression in hippocampus.Results:(1) In the model group,the escape latency(EL) was obviously longer than the sham operation group at each time point(P〈0.01).(2) Compared with the sham operation group,the number of Ng/P-GP positive neurons of hippocampal CA1 region was significantly decreased but that of GFAP/GLT-l positive cells increased(P〈0.01),and the expression of both the Ng/P-GP and GFAP/GLT-l were increased in model group(P〈 0.01);compared with the 15 d time point model group,the number of hippocampal Ng/P-GP positive neurons was highly decreased in 1 month time point model group but the number of GFAP/GLT-l positive cells and the expression of neuronal P-GP and glial GLT-l were increased in hippocampal CA1 region(P〈 0.01).(3) Compared with the sham operation group,integrated optical density(IOD) of Ng/P-GP and GFAP/GLT-1 positive cells in model group was significantly increased(P〈 0.01),which was much larger at 1 month time point than at 15 d in model group(P〈 0.01).Conclusion:The neuronal P-GP and glial GLT-l may be involved in the protection of hippocampal neurons in VD rats.
