摘 要:黄酮是桑根皮中的重要活性成分,建立高效、便捷提取桑根皮总黄酮的工艺技术,有利于对桑根皮药用活性成分的深度开发。基于超声波辅助乙醇提取桑根皮总黄酮的单因素试验结果,进一步采用Box-Behnken中心组合设计结合响应面分析法优化提取工艺参数,选择提取时间、原料质量浓度和乙醇体积分数作为考察因子,以桑根皮总黄酮得率作为响应值,利用Design Expert软件对数据进行回归分析并结合验证试验结果,确定桑根皮总黄酮的最佳提取工艺条件为提取时间1.6 h、乙醇体积分数80%、原料质量浓度0.04 g/m L。在此工艺条件下提取总黄酮的得率为3.63%,与预测总黄酮得率(3.71%)的相对误差为2.16%,说明优化的工艺条件具有一定的实用价值。Flavonoids are important active components of the mulberry root barks. Establishing an efficient and convenient extraction technology of total flavonoids from the root barks of mulberry will facilitate in-depth development of the medicinal active ingredients in mulberry root barks. Based on the single-factor experiment results from ultrasound-assisted ethanol extraction of total flavonoids from the root barks of mulberry, Box-Behnken central composite design combined with response surface analysis methodology (RSM) was further used to optimize the extraction technological parameters. By selecting extraction time, mass concentration of raw material and volume fraction of ethanol as the evaluation indexes, and the yield of total fiavonoids from mulberry root barks as response value, Design Expert software was applied to conduct regression analysis. Combined with validation tests, the optimized extraction process was determined as follows, extraction time 1.6 h, volume traction of ethanol 80%, and mass concentration of raw material 0.04 g/mE Under this optimal condition, the yield of total fiavonoids was 3.63%, having a relative error of 2. 16% with the predicted value (3.71%). The result indicated that the op- timized extraction process condition has certain practical value.
