人类外周血淋巴细胞培养的影响因素研究

摘　　要：将肘静脉血采集于含有肝素的一次性抗凝管中,采用RMPI1640培养基进行体外培养。通过对培养过程中血液用量、秋水仙素加入时间及浓度、低渗时间、滴片高度等条件的控制,得出血液用量为12-15滴,秋水仙素的浓度为100ug/ml时,能够得到形态完好,大小适中,分散均匀,轮廓清楚的中期染色体标本相。The elbow venous blood collected in disposable anticoagulant tube that containing heparin, using RMPI1640 medium and cultured in vitro. Through control dosage of blood , colchicines add time and concentra- tion, hypotonic time, drop height and other conditions, It is concluded that the amount of 12 to 15 drops of blood, the concentration of colchicines for 100ug/ml,can get good shape, size medium, distributed evenly, contour clear metaphase chromosome specimens.
