摘 要:目的探讨枕大池注入凝血酶(TM)后大鼠基底动脉蛋白酶激活受体-1(PAR-1)和肿瘤坏死因子-α(TNF-α)表达与基底动脉痉挛的关系。方法实验大鼠随机分为A(对照)组、B(单纯TM)组和C(全血+TM)组。A组大鼠枕大池内注入生理盐水,B组大鼠枕大池内注入TM 3U,C组大鼠枕大池注入自体血后再注入TM3U。术后3 d处死大鼠,测量基底动脉横截面积并检测PAR-1和TNF-α的表达。结果 1 B组较A组基底动脉横截面积小,PAR-1和TNF-α的表达高(P=0.000);2 C组较B组基底动脉横截面积小(P=0.002),TNF-α表达低(P=0.037),PAR-1表达差异不显著(P=0.42)。结论 TM可致血管缩窄、PAR-1和TNF-α的表达增高,并受SAH存在与否的影响。Objective The study aims to probe into the relationship between the expressions of proteinase- activated receptor-1 (PAR-l) and tumor necrosis factor-α (TNF-α) and vasospasm of basilar arterial in vivo after injection of thrombin (TIM) on basilar artery. Methods A total of 18 rats were divided into three groups: group A (control), group B (pure TM) and group C (TM + SAH) randomly. In group A, the rats were performed sham- operation with the injection of normal saline; in group B, the TM (3U) was injected into the animals'subarachnoid space via cisterna magna; in group C, the animals were all SAH models, and the TM (3U) was injected into the subarachnoid space after the injection of blood. Three days after operation, animals were killed and the basilar arteries of rats was used to detect the expressions of PAR-1 and TNF-α by immunohistochemistiy, and then the mean optical density (MOD) of PAR-1 and TNF-α were taken as the semiquantitative indicators for statistical analysis. Results Compared with group A, the rats'basilar artery lumen cross-sectional area of group B was smaller, and the MOD of PAR-1 and TNF-α of basilar artery were higher than those of group A (P =0. 0130) ; the basilar artery huron cross-sectional area of group C were smaller than that of group B (P 〈0. 05); meanwhile, the MOD of TNF-ct of group C was lower than that of group B (P 〈0.05); but there was no significant difference in MOD value of PAR-1 between group B and C (P 〉0.05). Conclusion The TM in CSF will lead to cerebral vasospasm (CVS) and higher expressions of PAR-1 and TNF-α, and its effect is associated with the presence or absence of SAH.
