摘  要:以黄芩悬浮细胞为材料研究其生长过程中蛋白质含量变化规律及与黄芩苷次生合成之间的关系. 结果表明,在黄芩悬浮细胞生长周期(21 d)内,蛋白质含量的高低与黄芩苷的积累呈正相关.在黄芩悬浮细胞生长周期的前6 d,悬浮细胞处于潜伏期,蛋白质含量低水平表达,黄芩苷合成缓慢; 6~18 d,悬浮细胞处于生长期,蛋白质的含量高水平表达,合成速率较快,这期间黄芩苷也迅速合成; 18~21 d,悬浮细胞进入稳定期,此时蛋白质含量处于稳定期,黄芩苷合成也几乎停止. 同时采用L9(34)正交试验研究培养基中6-BA、碳源、氮源、pH的配比对蛋白质含量表达的影响,结果表明碳磷源(NH4+)对蛋白质的表达影响最大.Taking the scutellaria suspensioncells as the material, the relationship between the protein content and baicalin secondary synthesis in the growth process of the scutellaria suspension cells was investigated. The results showed that during the growth cycle (21 days) of the scutellaria suspension cells, the content of protein and the accumulation of baicalin was positively correlated. During the former 6 days of the growth cycle of the scutellaria suspension cells, the skullcap cells was in incubation period, the content of the protein express at a low level and the synthesis of baicalin was slow. During 6 to 18 days, the cells was in its growing phase, the content of protein expressed at high levels, at the same time baicalin rapidly synthetised. During 18 to 21 days, the cells was in its stable period, the content of baicalin was also in its stable period, the synthesis of baicalin almost stopped, meantime, L9(34) orthogonal table was used to optimize the ratio of medium 6-BA, carbon sources, nitrogen sources and pH, it was conducted that carbon sources had the maxium influence on the protein expression.
