摘  要:目的探究血卟啉单甲醚(hematoporphyrin monomethyl ether,HMME)介导的光动力疗法(photodynamic therapy,PDT)对人食管癌生长的抑制作用。方法以人食管癌ECA-109细胞制备出裸小鼠人食管癌移植肿瘤模型,经裸小鼠尾静脉注入HMME液(20 mg/kg),光敏化2 h后用半导体激光仪(波长为630 nm)垂直照射肿瘤(能量密度4 J/cm2),荷瘤对照组、单纯激光组、单纯HMME组和HMME-PDT治疗组分别治疗后,测量裸鼠肿瘤大小并计算肿瘤体积,末次测量后切除肿瘤经HE染色后行病理形态学观察。结果 HMME-PDT治疗组肿瘤体积明显小于其他三组,差异有统计学意义(P<0.05),经HMME-PDT治疗后肿瘤细胞大片凝固性坏死,HMME-PDT治疗组平均生存时间(35 d)长于对照组(24 d)。结论 HMME-PDT治疗能明显抑制人食管癌裸小鼠移植瘤的生长,并可延长荷瘤裸小鼠的生存期限,而单纯激光组和单纯HMME组不能明显抑制裸鼠移植瘤的生长。Objective To explore the effects of hematoporphyrin monomethyl ether mediated photodynamic therapy (HMME-PDT) on ECA-109 human esophageal cancer. Methods ECA-109 human esophageal cancer xenografts were established in nude mice. The animals bearing xenografts were treated with 20 mg/kg weight HMME and 2 hours later were irradiated with 4 J/cm2 light from the Semiconductor laser with a wavelength of 630 nm. The tumor sizes in nude mice (tumor-bearing control group, the laser group, the HMME group, HMME-PDT treatment group) were measured after treatment and then the tumor volumes were calculated, the tumors were removed after the last measure, then the tumor tissues were stained with HE. Results The volume in HMME-PDT treatment group was obviously smaller than the other three groups, there was obviously significant difference (P 〈 0.05 ). After HMME-PDT treatment, tumor cells showed large areas of coagulation necrosis. The mean survival time was longer than that of tumor-bearing control group. Conclusion HMME-PDT treatment can obviously inhibit the growth of human esophageal cancer xenograft and extend the lifespan of tumor-bearing nude mice. The laser group and the HMME group can not obviously inhibit the growth of human esophageal cancer xenograft.
