摘  要:目的探讨WHO引进的Vero细胞对狂犬病病毒(rabies virus,RV)aGV株的易感性。方法将RV aGV按MOI 0.01-0.1感染Vero细胞主代和工作代,显微镜下观察细胞形态;免疫荧光法观察感染24、48、72和96 h细胞内病毒的增殖情况;连续培养15 d,每天取上清液,采用免疫荧光法检测病毒滴度。结果 RV感染Vero细胞可致细胞圆缩病变,病毒感染第7天,病变率约为20%;RV感染Vero细胞主代和工作代24 h单个细胞内可见特异性荧光灶,48 h 20%-30%细胞内可见荧光灶,72 h 60%-80%细胞内可见荧光灶,96 h 90%-100%细胞内可见荧光灶;病毒感染后2-8 d,上清液中检测到的RV滴度均在103-108 FFU/ml之间波动,均为第8天达到高峰,达5.0×108FFU/ml以上。结论 WHO引进的Vero细胞对RV aGV株有较强的易感性,可应用于狂犬病疫苗的规模化生产。Objective To investigate the sensitivity of Vero cells introduced by WHO to rabies virus(RV) aGV strain.Methods Vero cells were infected with the master and working seeds of aGV strain at MOIs of 0. 01 - 0. 1, and observed for morphology under microscope. The proliferations of virus in cells 24, 48, 72 and 96 h after infection were determined by IFA. The virus was cultured for 15 d, and the supernatant was collected every day and determined for virus titer by IFA. Results Infection with RV caused cytopathic effect(CPE). The CPE rate on day 7 after infection was about 20%.Specific fluorescent foci were observed in single Vero cell 24 h, in 20% - 30% of cells 48 h, in 60% - 80% of cells 72 h,and in 90% - 100% of cells 96 h, after infection with master and working seeds of RV. The RV titer in supernatant was103- 108 FFU / ml 2 - 8 d, and reached a peak value of more than 5. 0 × 108 FFU / ml on day 8, after infection.Conclusion The Vero cells from WHO was highly susceptible to RV aGV strain, which might be used for the large scale production of rabies vaccine.
