摘 要:目的:观察多烯紫杉醇对人乳腺癌细胞T47D增殖与凋亡的影响及其机制研究。方法:用不同浓度(0、5、10、20nmol·L-1)多烯紫杉醇作用于人乳腺癌细胞T47D后,采用MTT法观察多烯紫杉醇对人乳腺癌细胞T47D增殖的影响,并使用流式细胞术及Hoechst33258染色检测多烯紫杉醇对T47D细胞凋亡的影响,采用RT-PCR检测T47D细胞bcl-2mRNA表达情况。结果:经多烯紫杉醇处理后,MTT结果显示T47D细胞生长抑制率显著上升(P<0.05),流式细胞术检测显示T47D细胞凋亡率显著升高(P<0.05),并且Hoechst33258染色检测T47D细胞呈凋亡状态。此外,RT-PCR检测T47D细胞bcl-2mRNA表达水平降低(P<0.05)。结论:多烯紫杉醇通过诱导凋亡能抑制人乳腺癌细胞T47D的增殖,这可能与其下调bcl-2mRNA表达有关。Objective:To study the effects and the mechanisms of docetaxel on the proliferation and apoptosis of human breast cancer cells T47 D. Methods:Human breast cancer cells T47 D was exposed to different doses of docetaxel(0,5,10 and20nmol·L-1).MTT assay was applied to determine the effects of docetaxel on proliferation and apoptosis of human breast cancer cells T47 D,while apoptosis was assessed by flow cytometry and Hoechst33258 staining.Next,the expression level of bcl-2mRNA in T47 Dcell was analyzed by RT-PCR.Results:After docetaxel treatment,the proliferation of T47 Dcell was significantly inhibited(P〈0.05).The increased apoptotic rates by docetaxel was found in T47 Dcell by flow cytometry(P〈0.05),and docetaxel-treated T47 Dcell exhibited typical apoptotic morphology.Furthermore,docetaxel resulted in reduced expression of bcl-2mRNA in T47 D cells(P〈0.05).Conclusions:Docetaxel could inhibit the growth of human breast cancer cell T47 Dby induction of apoptosis,which may be related with inhibition of bcl-2mRNA expression.
