摘  要:目的探讨机械性张应力作用对成牙骨质细胞增殖及分化功能的影响。方法采用流式细胞术及RT-PCR对接受了不同时间点张应力作用的成牙骨质细胞OCCM30进行细胞周期活性变化的检测及成骨相关矿化基因m RNA的检测。结果各时间点间加力组增殖指数(PI)与0h相比差异均具统计学意义;3h,6h,12h,18h加力组与各自的对照组差异具有统计学意义;周期性张应力可规律性促进OCCM30相关矿化基因Runx2、Osterix、ALP、ColⅠm RNA的表达。结论周期性张应力刺激可引起成牙骨质细胞的细胞周期和增殖活性的变化,可促进OCCM30相关矿化基因Runx2、Osterix、ALP、ColⅠm RNA的表达。Objective To investigate the influence of tension stress on cementoblast proliferation and differentiation. Methods Cementoblast OCCM30 were cultured subjected to mechanical tension stress for 0h, 1h, 3h, 6h,9h, 12 h, 18 h and 24 h. The flow cytometry and RT-PCR were used to examine the cell cycle and proliferation activity and mineralization related m RNA genes after loading at each time point. Results There was statistical significant difference between stress loading group and stress loading free group. There was also statistical significant difference between stress loading groups and their control groups for 3h, 6h, 12 h, 18 h. Tension stress can promote the expression of m RNA of Runx2、 Osterix、 ALP、 Col Ⅰ statistically significant. Conclusion Different loading time can induced different proliferation activity of cementoblast. Tension stress can regulate the proliferation and differentiation activity of cementoblast of OCCM30.
