摘 要:目的:探讨小鼠诱导性多能干细胞(i PS,induced pluripotent stem cell)体内向牙源性细胞分化的潜能及适宜的诱导微环境。方法:经成釉细胞无血清条件培养液(ASF-CM)以及加入BMP4的ASF-CM(ASF-BMP4)不同微环境体外诱导的i PS细胞,分别移植入小鼠肾被膜内,H E染色及免疫组织荧光检测相关指标的表达。结果:ASF-CM组移植物HE染色可见上皮样结构生成,免疫组织荧光检测AMBN表达阳性;ASF-BMP4组可见上皮样和牙本质样结构生成,免疫组织荧光检测AMBN和DSP表达阳性。结论:ASF-BMP4提供了一个适宜的成牙微环境,能够促进i PS细胞体内形成牙源性上皮样和牙本质样结构。Objective: To study the possibility and conditions of differentiation of mouse induced pluripotent stem(i PS)cells into odontogenic cells in vivo. Methods: An in vivo transplantation assay was performed to further investigate the in vivo differentiation capacity of mouse i PS cells. Aggregates of i PS cultured in ASF-CM or in ASF-BMP4 for 14 days with CBB were implanted into the renal capsule of C57 mice. Haematoxylin and eosin(HE) and immunofluorescent staining were used to detected odontogenesis-related protein expression of AMBNand DSP. Results: ASF-CM-treated mouse i PS cells only generated ameloblast-like tissues. The expression of AMBN was found positive by immunofluorescent staining.ASF-BMP4-treated mouse i PS cells formed both ameloblast-like and odontoblast-like structures. The expression of AMBN and DSP were positive. Conclusion: These results suggest that mouse i PS cells can be induced by ASF-BMP4 to differentiate into ameloblast-like and odontoblast-like cells in vivo. ASF-BMP4 provides a suitable microenvironment for teeth regeneration.
