摘 要:目的研究葛根-黄连药对提取物对α-葡萄糖苷酶活性及大鼠餐后高血糖的抑制作用。方法提取大鼠小肠α-葡萄糖苷酶,以蔗糖为底物,用葡萄糖氧化酶法研究葛根-黄连药对、葛根和黄连单味药材提取物对α-葡萄糖苷酶的抑制活性。以蔗糖诱导的餐后高血糖大鼠为模型,测定灌胃给予葛根-黄连药对提取物后的血糖值,研究葛根-黄连药对对大鼠餐后高血糖的抑制作用。结果葛根-黄连药对、葛根和黄连提取物对α-葡萄糖苷酶的抑制作用呈剂量依赖性,IC50分别为2.439、3.700和5.730 g·L^(-1),葛根-黄连药对提取物对该酶的抑制作用最强;高剂量组的葛根-黄连药对提取物(400 mg·kg^(-1))可显著降低大鼠进食蔗糖后的血糖值。结论葛根-黄连药对提取物对α葡萄糖苷酶的抑制作用显著强于单味药材提取物,并可显著缓解大鼠餐后血糖升高。Objective To investigate the inhibitory effects of Puerariae lobatae Radix-Rhizoma coptis herb couple( PR) extract on α-glucosidase and high postprandial blood glucose levels in rats. Methods The α-glucosidase of rat small intestine was extracted in vitro,the inhibitory effects of the extract of PR,Puerariae lobatae Radix( PLR) and Rhizoma coptis( RC) were studied by the method of glucose oxidase using the sucrose as the substrate. The rats model high postprandial blood glucose were induced by sucrose,the blood glucose was measured after intragastic administration of PR extract in order to investigated the inhibitory effect of PR extract on high postprandial blood glucose in rats. Results The inhibitory effect of PR,PLR and RC extract displayed dose-dependent manner with IC50 values 2. 439,3. 700 and 5. 730 g·L^(-1) respectively,the PR extract had the strongest inhibitory activity on α-glucosidase. High dose group of PR extract(400 mg·kg^(-1)) could decrease the blood glucose significantly in rats after sucrose administration. Conclusions Extract of PR has stronger inhibitory effect on α-glucosidase than that of single herbs and can alleviate the high postprandial blood glucose significantly.
