乳腺癌组织Ki-67与p53的表达特征及与预后的关系

摘　　要：目的分析乳腺癌组织Ki-67与p53的表达特征及其与预后的关系。方法选取70例行乳腺癌根治手术的乳腺肿瘤患者癌组织标本,采用免疫组织化学(Immunohistochemistry,IHC)SP二步染色法检测乳腺癌组织(研究组)及其癌旁组织(对照组)中Ki-67和p53的表达;结合术后随访资料,比较5年内复发转移患者与无复发转移患者的Ki-67、p53表达水平。结果研究组Ki-67、p53蛋白阳性率均显著高于对照组(均P<0.01);复发转移组的Ki-67、p53阳性细胞表达率均显著高于无复发转移组(均P<0.05)。乳腺癌患者的生存率与癌组织中Ki-67、p53阳性细胞数显著正相关(r=0.712,P<0.05)。结论乳腺癌组织中Ki-67、p53表达水平与乳腺癌复发转移及不良预后相关。Objective To find out the pattern of the expression of Ki-67 and P53 in breast cancer tissue and its correlation to prognosis. Method Specimen of breast cancer tissue was firstly obtained from 70 breast cancer patients having undergone. Immunohistochemistry （IHC） SPtwo-step staining was then used to detect the expression of Ki-67 and P53 in the cancer tissue （the control subject） and the expression of Ki-67 and P53 in the tissue （ the experimental subject） adjacent to the cancer. Based on data obtained from follow-up, the expression of Ki-67 and P53 of cases of recurrence within 5 years was compared with that of cases of whom recurrence was not found. Findings The Ki-67 and p53 protein positive rate of the control subject was significantly higher than that of the experimental subject. The difference was statistically significant （X2 =6. 132, P = 0. 000; X2 = 5. 973, P = 0. 000）. The recurrence group＇s Ki-67 and p53 positive cells number was significantly higher than those of the group without recurrence. The difference between the two groups was significant （X2 = 2. 542, P = 0. 016; X2 = 2. 610 ,P = 0. 008 ）. Survival rate of breast cancer patients was positively correlated to the number of positive cells of Ki-67 and P53. Conclusion The level of expression of Ki-67 and P53 is positively correlated to the recurrence, metastasis and poor prognosis of cases of breast cancer.
