摘 要:目的观察低频强声作用后大鼠海马回细胞形态、分布及凋亡的变化,探讨其可能机制。方法 SD大鼠随机分为:对照组8只和实验组40只。实验组:100Hz,140d B低频强声,1h/d,连续作用20d,在1d、10d、20d及停止作用后10d、20d处理大鼠,每次8只。采用HE染色观察海马回细胞形态和分布,TUNEL法检测海马回细胞凋亡,免疫组化检测海马回NSE和GFAP的表达。结果 1作用10d可见海马回细胞排列稍疏松紊乱,作用20d细胞排列疏松紊乱明显,而停止作用后10d细胞疏松紊乱现象较作用20d有所改善,停止作用后20d未进一步改善。2随着低频强声作用时间延长,海马回TUNEL阳性细胞率呈升高趋势,停止低频强声作用后有所回落,但仍高于对照组。3海马回NSE阳性细胞数量随着低频强声作用时间延长呈减少趋势,停止低频强声作用后有所增加,但仍少于对照组,而GFAP阳性细胞数量无明显变化。结论长时间低频强声作用可导致海马回神经元数量显著减少,停止作用后神经元数量部分恢复。Objective To observe the morphology,structure and cells apoptosis of hippocampal gyrus in rats exposed to strong lowfrequent noise and explore its mechanism. Methods SD rats were exposed to 100 Hz,140d B strong low-frequency noise 1h / day for20 days. At 1th、10th、20th days during exposure and 10th、20th after stopping exposure respectively,the brain tissues of eight rats were detected by using HE staining,immune-histo-chemistry and TUNEL method. Results( 1) Cells of hippocampal gyrus were turning sparse at the 10 thday of exposure to strong low-frequency noise and at the 20 thday of exposure these changes were worse. When stopping exposure,the sparsity of rat hippocampus cells was recovered partly after 10 days,but little improvement hereafter.( 2) TUNEL method showed the percentage of positive cell of hippocampal gyrus increased sharply following the adding of exposure time to strong low-frequency noise. It decreased after stopping exposure,but still higher than that of the control group.( 3) ICC staining showed the NSE positive cells reduced sharply related to the exposure time prolonged to strong low-frequency noise and it turned better after stopping exposure,but still less than that of the control group. The GFAP positive cells had no change in the course of the test.Conclusion The number of neurons in hippocampal gyrus decreased significantly after long-time exposure to strong low-frequency noise,however,they could recover partly after stopping exposure.
