摘 要:为探讨蝙蝠葛碱(DAU)对阿尔茨海默病(AD)大鼠海马区乙酰胆碱酯酶(AChE)和胆碱乙酰化酶(ChAT)的影响,采用海马立体定向注射凝聚态Aβ1-42制备AD大鼠模型。DAU高、中、低3个剂量组进行灌胃给药干预,给药4w后,处死大鼠,取海马区组织,采用免疫组织化学法检测分析AChE和ChAT的表达。与对照组比较,模型组AChE的蛋白表达明显增高(P<0.05),而ChAT的蛋白表达明显减少(P<0.05);与模型组比较,治疗组,DAU高、中、低各剂量组中AChE表达均明显降低(P<0.05),而ChAT表达明显升高(P<0.05)。说明DAU通过下调大鼠海马AchE表达和上调ChAT表达发挥抗AD作用。In order to investigate the effect of DAU on AChE and ChAT in hippocampus of rats with Alzheimer's disease AD, the model of AD rat was prepared by three dimensional A beta 1-42. DAU, middle and low dose groups were administered orally to the rats. After administration of4 W, the rats were killed and the ACh E and Ch AT expression was detected by immunohistochemistry. Compared with the control group, the protein expression of ACh E in model group was significantly increased(P〈0.05), and the expression of Ch AT protein was significantly decreased(P〈0.05). The expression of ACh E in the treatment group was significantly lower than that in the treatment group DAU, while the expression of Ch AT was significantly increased(P〈0.05). DAU shows that expression and Ch AT expression of Ach E in the hippocampus of rats play an anti AD effect.
