人羊膜上皮细胞上清液对人肝癌BEL-7402细胞的体外抗肿瘤作用

摘　　要：目的探讨人羊膜上皮细胞(h AECs)上清液对体外培养的肝癌细胞系BEL-7402细胞迁移、增殖与凋亡的影响。方法 BEL-7402细胞随机分为5组,其中对照组不加h AECs上清液,其余4组加入体积分数分别为6.25%、12.5%、25%和50%的h AECs上清液作用24 h后,通过划痕实验、MTT试验和流式细胞术检测BEL-7402细胞迁移、增殖和凋亡;Western blot法检测凋亡相关蛋白的表达量。结果 h AECs上清液能剂量依赖性地抑制BEL-7402细胞迁移、增殖并诱导凋亡(P<0.05);25%和50%h AECs上清液组BEL-7402细胞caspase-3和caspase-8蛋白的切割片断蛋白定量明显高于对照组(P<0.05)。结论 h AECs上清液对体外培养的BEL-7402细胞具有一定的抗肿瘤作用。Objective To investigate the migration,proliferation and cell apoptosis effects of human amniotic epithelial cells（ hAECs） supernatant on human hepatoma BEL-7402 cells. Methods BEL-7402 cells were randomly divided into five groups. The control group contained no hAECs supernatant,while the other four treatment groups were incubated with different volume fractions of hAECs supernatant for 24 hours. The scratch assay,MTT assay and flow cytometry were used to detect the migration,proliferation and apoptosis of BEL-7402 cells. The expression of apoptosis-related proteins was detected by Western blot. Results hAECs supernatant dose-dependently inhibited the migration and proliferation of BEL-7402 cells and induced apoptosis（ P〈0. 05）. The expression of cleaved caspase-3 and cleaved caspase-8 of BEL-7402 cells in 25 % and 50 % hAECs supernatant groups were higher than those in control group（ P〈0. 05）. Conclusions hAECs supernatant has some antitumor effects on BEL-7402 cells in vitro.
