STAT3及p-STAT3在皮肤鳞状细胞癌中的表达及其与鳞癌临床病理特征的关系

摘　　要：目的探讨信号传导及转录活化因子3(signal transducer and activator of transcription 3,STAT3)和磷酸STAT3(pSTAT3)在皮肤鳞状细胞癌中的转录表达,以及p-STAT3表达与皮肤鳞状细胞癌临床病理特征的关系。方法收集36例皮肤鳞状细胞癌患者癌组织及癌旁正常组织,RT-PCR检测STAT3转录情况,免疫组化方法检测STAT3和p-STAT3蛋白表达水平;对比分析p-STAT3阳性表达组患者和阴性表达组患者年龄、性别、肿瘤大小、组织病理分化的差异。结果癌组织中STAT3转录水平高于对应癌旁正常组织(P<0.01),癌组织中STAT3和p-STAT3蛋白阳性表达率均高于癌旁正常组织(P<0.01)。癌组织p-STAT3阳性表达组与阴性表达组比较,患者在肿瘤大小、组织病理分化的差异有统计学意义。结论皮肤鳞状细胞癌中存在STAT3的异常表达和活化,p-STAT3是评估患者预后可能指标之一。Objective To investigate the expression of signal transducer and activator of transcription 3（ STAT3） and p-STAT3 signal transducer and activator of transcription 3（ p-STAT3） in human squamous carcinoma cells（ SCC） and to explore the relationship between p-STAT3 expression and clinicopathological characteristics of SCC. Methods The transcription of STAT3 in SCC tissues and paracancerous tissues from 36 SCC patients were detected by reverse transcription PCR. The protein expression levels of both STAT3 and p-STAT3 were detected by immunohistochemistry. Patient age,gender,tumor size,and pathological grade were compared between p-STAT3 positive group and p-STAT3 negative group. Results The level of STAT3 mRNA was significantly higher in SCC tissues than that in paracancerous tissues（ P 〈 0. 01）. The positive expression rates of STAT3 protein and p-STAT3 protein in SCC tissues were higher than those in paracancerous tissues（ P 〈 0. 01）. The tumor size and pathological grade were significantly different between pSTAT3 positive group and p-STAT3 negative group（ P 〈 0. 01）. Conclusion The abnormal expression and activation of STAT3 exist in SCC,and p-STAT3 may be a potential tumor marker to predict the prognosis of SCC.
